肿瘤易感基因101的裸鼠荷瘤实验

摘要 目的探讨胃癌细胞SGC7901中高表达的肿瘤易感基因101(TSG101)对肿瘤增殖的作用及机制。方法将不同TSG101表达水平的胃癌细胞亚系接种于裸鼠体内,观察移植瘤生长状况及相关因子的表达。结果 TSG101转染组裸鼠的移植瘤体积明显高于空质粒组与未转染组(P<0.05)。TSG101转染组瘤组织切片中血管内皮细胞生长因子(VEGF)表达明显高于空质粒组和未转染组(P<0.05)。结论 SGC7901细胞中高表达的TSG101可能通过上调VEGF的表达增强胃癌细胞增殖。 Objective To explore the effect and mechanism of tumor susceptibility gene 101(TSG101) of highly expressed gastric cancer SGC7901 cell on tumor proliferation. Methods Cells were divided into non-transfection group, empty plasmid transfection group and TSG101 eukaryotic expression plasmid transfection group(the TSG101 transfection group). Then observed the growth conditions of transplantation tumor and the expression of correlation factor. Results The expression of VEGF of the TSG101 transfection group of tumor tissue in nude mice was significantly higher than that of the non-transfection group and the empty plasmid transfection group(P〈0.05). The transplantation tumor volume of the TSG101 transfection group of tumor tissue in nude mice was significantly higher than that of the non-transfection group and the empty plasmid transfection group(P〈0.05). Conclusion The high-expression of TSG101 in SGC7901 cells may enhance the proliferation of gastric cancer by up-regulate the expression of VEGF.
