摘要
采用细胞病变效应(CPE)观察和活细胞染色法(MTS),研究了银黄等几种天然植物药物在细胞培养模型(体外培养的锦鲤鳍条细胞)上抑制锦鲤疱疹病毒(KHV)复制的效果。药物安全浓度试验结果表明,银黄、黄芪多糖、板蓝根、炎琥宁以及穿心莲对锦鲤鳍条组织细胞的安全浓度分别为15.63、2.5、31.25、31.25 mg/mL和6.25 mg/mL。在先给药后感染试验组,银黄、黄芪多糖、板蓝根和炎琥宁的浓度分别为安全浓度时,对KHV有明显的抑制效果,治疗指数分别为22.88±3.26、21.07±4.86、24.76±3.24和18.20±2.78;穿心莲抑制病毒复制的效果不明显。在先感染后给药试验组,银黄、板蓝根和炎琥宁浓度分别在安全浓度时,对KHV有显著的抑制作用,治疗指数分别为33.21±7.71、21.99±3.12和15.91±3.23。使用天然植物药物处理病毒后再感染细胞,仅观察到板蓝根在浓度为31.25 mg/mL体外对KHV有较弱的直接杀灭作用,治疗指数为5.7±1.62,而银黄、黄芪多糖、穿心莲和炎琥宁没有直接杀灭KHV的效果。
By using the observation of cytopathic effect( CPE) and the staining assay of live cells with MTS,herbal drugs were screened and evaluated for the inhibition of replication of koi herpesvirus( KHV) in Koi- Fin cell line. The safety tests of drugs to cells showed that Yinhuang,Astragalus polysaccharide,Radix Isatidis,Herbal Andrographitis and Andrographolide were 15. 63 mg / mL,2. 5 mg / mL,31. 25 mg / mL,31. 25 mg / mL and 6. 25 mg / mL,respectively. In the tested group of drugs added prior to infection, while the concentrations of Yinhuang, Astragalus polysaccharide, R. Isatidis and Herbal Andrographitis were 15. 63 mg / mL,2. 5 mg / mL,31. 25 mg / mL and 6. 25 mg / mL,significant inhibition of KHV replication were observed,the treatment index were 22. 88 ± 3. 26,21. 07 ± 4. 86,24. 76 ± 3. 24 and 18. 20 ± 2. 78,respectively,but the Andrographolide is of no apparent inhibition. In the tested group that the infection was prior to drug addition,at the concentrations of Yinhuang,R. Isatidis and Herbal Andrographitis of 15. 63 mg / mL,31. 25 mg / mL and 6. 25 mg / mL,the significant effects in inhibition of KHV replication were achieved,the treatment index were 33. 21 ± 7. 71,21. 99 ± 3. 12 and 15. 91 ± 3. 23,respectively,while the Astragalus polysaccharide and Andrographolide had no apparent effects. In the tested group of co- incubation of virus and drugs before infection,at the concentrations of R. Isatidis of 31. 25 mg / mL,low efficacy was observed in inhibiting KHV replication,the effect of treatment index were5. 7 ± 1. 62,while the other drugs had no effects.
