摘要
目的建立一种LC-MS/MS的分析方法同时检测复方祖师麻提取物(祖师麻与甘草质量比3∶2)中8种成分(瑞香素、西瑞香素、7-OH香豆素、甘草酸、甘草苷、异甘草苷、甘草素和异甘草素),并应用此方法考察两者配伍前后活性成分提取率的变化情况。方法 Hypersil Gold-C18色谱柱(50 mm×2.1 mm,1.9μm),乙腈-0.05%甲酸水梯度洗脱,体积流量为0.3 mL/min,柱温为35℃。质谱条件为采用负离子模式,以SRM方式进行定量。结果甘草配伍祖师麻后,祖师麻中3种活性成分(瑞香素、西瑞香素和7-OH香豆素)的量均有一定程度的增加,其中尤以瑞香素的增加最为显著;祖师麻降低甘草中苷类成分(甘草酸、甘草苷和异甘草苷)溶出,而对苷元(甘草素和异甘草素)的溶出稍有提高。结论该分析方法能快速、特异、灵敏并可重复检测祖师麻和甘草中的主要活性成分,两者配伍对8种成分提取率的影响并不相同。
AIM To use LC-MS /MS method to simultaneously analyse eight active components( daphnetin, daphnoretin,7-hydroxycoumarin,glycyrrhizin,liquiritin,isoliquiritin,liquiritigenin and isoliquiritigenin) extracted from combination of Zushima( Daphne giraldii Nitsche.) and Gancao( Glycyrrhizae Radix et Rhizoma)( ratio of 3 ∶ 2) and their mutual solubility. METHODS The analytes were seperated by Hypersil Gold-C18( 50 mm × 2. 1 mm,1. 9 μm) at 35 ℃ and acetonitrile-0. 05% formic acid used as the mobile phase with gradient elution on the rate of 0. 3 mL / min. A negative ionization selective reaction monitoring model and electrospray ionization source was applied to the analysis method. RESULTS The separption efficient peaks presented a good linearity. After Zushima combined with Gancao,the concentrations of three components in Zushima( daphnetin,daphnoretin and 7-hydroxycoumarin) increased to some extent,especially for daphnetin. The concentrations of glycosides( glycyrrhizin,liquiritin and isoliquiritin) in Gancao decreased while that of aglycones( liquiritigenin and isoliquiritigenin) increased. CONCLUSION The method is rapid,simple and accurate,which can be used to simultaneously detect the concentrations of the main components in Zushima and Gancao,and the influence of compatibility on their extraction rates is not the same.
