摘要
目的:观察祖师麻注射液对巨噬细胞分泌血管内皮生长因子(VEGF)的影响并探讨其机制。方法:将巨噬细胞RAW264.7与脂多糖及不同浓度的祖师麻注射液共同培养,MTT法检测细胞的增殖,ELISA检测细胞培养液中VEGF的含量,Wstern blot检测缺氧诱导因子-1a(HIF-1a)的表达。结果:随着祖师麻浓度从218.7mg/ml下降至0.3mg/ml,其对RAW264.7细胞增殖的抑制率也从34.04%下降至2.28%;经218.7mg/ml、72.9mg/ml、24.3mg/ml祖师麻处理后,RAW264.7细胞分泌的VEGF从1895±155pg/ml分别减少至1029±175pg/ml、1575±186pg/ml、1550±217pg/ml,同时细胞内HIF-1a的表达也明显降低。结论:祖师麻注射液可抑制巨噬细胞RAW264.7分泌VEGF,其机理与抑制HIF-1a的表达密切相关。
Objective: To investigate the effects of Zushima injection(ZSM)on the expression of vascular endothelial growth factor(VEGF) in macrophages and explore the mechanism. Methods: RAW264.7 cells were co - incubated with lipopolysaccharide ( LPS ) and various doses of ZSM. MTT assay was used to identify the proliferation of cells. The secretion of VEGF quantified by ELISA test and the expression HIF - 1 a was analyzed by western blot assay. Results: When the doses of ZSM reduced from 218.7mg,/rrd to 0.3mg/ml the inhibition rate of ZSM on RAW264.7 cells decreased from 34.04% to 2.28%. After added ZSM at the doses of 218.7mg/ml, 72.9mg/ml, 24.3mg/ml to cells, the secretion of VEGF decreased from 1895 :l: 155 pg/ml to 1029 ± 175 pg/ml, 1575 ± 186 pg/ml, 1550 ± 217 pg/ml separately, and at the same time, the expression of HIF -1 a also decreased markedly. Conclusion: ZSM can remarkably reduce the secretion of VEGF, and its mechanism might be associated with inhibiting the expression of HIF - 1a.
