摘要
目的 建立高效液相色谱法(HPLC)同时测定无梗五加中7个有机酸(绿原酸,新绿原酸,咖啡酸,1,3-二咖啡酰奎宁酸,异绿原酸B,异绿原酸A,异绿原酸C)含量的方法.方法 采用Hypersil ODS2 C18(250 mm×4.6 mm,5μm)色谱柱,柱温室温,以0.1%甲酸乙腈-0.1%甲酸水溶液进行梯度洗脱,流速1.0 mL/min,检测波长324 nm.结果 7个有机酸的线性范围分别为20.5 ~328、1.99 ~31.8、2.43 ~38.9、2.44~39.0、2.52~40.3、7.25~116、2.42 ~ 38.7 mg/L,平均加样回收率均在95.2% ~ 103.9%范围内.结论 该方法快速简便、精密度好、灵敏度高,为控制无梗五加药材质量及与五加属的其他药材进行对比研究提供了方法.
Objective To establish a HPLC method for simultaneous determination of 7 kinds of organic acid in Acanthopanax sessiliflorus (chlorogenic acid, neochlorogenic acid, caffeic acid, 1,3-dicaffeoylquinic acid, 3,4-dicaffeoylquinic acid, 3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid ). Methods The HPLC procedure was performed on the chromatographic column of Hypersil ODS2 C18 (250 ×4.6 mm, 5 μm) at room temperature, and mobile phase was 0.1% formic acid in acetonitrile -0.1% formic acid in water. The flow velocity was 1.0 mL/min and detection wavelength was 324 nm. Results All 7 kinds of organic acid showed good linear relationship respectively at range of 20.5-328 μg/mL, 1.99-31.8μg/mL, 2.43-38.9μg/mL, 2.44-39.0μg/mL, 2.52-40.3 μg/mL, 7.25-116 mg/L and 2.42-38.7μg/mL. The average recoveries of them were within a range from 95.2% to 103.9%. Conclusion The method is quick, simple, accurate and sensitive, which can be used as a method for controlling the quality of Acanthopanax sessiliflorus and other Acanthopanax materials.
