摘要
α2巨球蛋白(α2-macroglobulin,α2M)是血浆中含量丰富的大分子糖蛋白,是一种广谱的蛋白酶抑制剂,主要通过调节内源或外源性蛋白酶广泛参与许多重要的生理病理过程。α2M还通过与细胞因子、生长因子和激素等分子结合调节它们在体内的分布、半衰期和生物学活性。研究发现α2M具有广泛的应用前景:α2M对辐射损伤、脓毒症、阿尔茨海默病等疾病具有很好的保护作用。此外,α2M还可以作为药物传递的载体和疫苗佐剂,并且可以作为许多疾病诊断和预后的生物标志物。本文主要综述α2M在应用方面的新进展。
α2-macroglobulin (α2M) is a large molecular weight glycoprotein which is abundant in the plasma of vertebrates and invertebrates. As a broad-spectrum protease inhibitor, α2M participates in many important physiological and pathological processes. It can also bind cytokines, growth factors, and hormones and regulates their distribution, half-life period, and biological activity. Recent studies have showed that α2M exerts protective role against acute radiation syndrome, septic shock, Alzheimer Disease, etc. Furthermore, α2M can serve as drug delivery vehicle and vaccine adjuvant. It can also be used as an early biomarker in diagnosis and prognosis of many diseases. Therefore, α2M has a broad application prospect. Here, we review the research progress on α2M applications.
