摘要
以元蘑为原料,采用超声波辅助技术,考察超声提取时间、乙醇体积分数、超声提取功率、料液比对元蘑总三萜提取率的影响,以响应面法优化工艺条件;并利用4种大孔树脂进行纯化。结果表明,超声波辅助提取元蘑总三萜最佳提取工艺条件为:乙醇体积分数80%、超声提取功率350 W、料液比1︰26(g/mL)、超声提取时间30 min、超声提取温度40℃;在此条件下元蘑总三萜的提取率达3.06%;NKA-9树脂为最佳树脂,经纯化的元蘑总三萜纯度可达到9.8%。
Ultrasound-assisted extraction of total triterpenoids from Hohenbuehelia serot/na was investigated. Effect of ultrasonic extraction time, ethanol concentration, ultrasonic power and material-liquid ratio on extraction yield of total triterpenoids was investigated by single-factor experiment, and the response surface methodology was used to optimize the extraction parameters. Four kinds of commercial macroporous resins had been selected to purify total triterpenoids. The results showed that the optimum extraction condition of ultrasonic assisted exlraction was: ethanol concentration 80%, ultrasonic power 350 W, ratio of solid to liquid 1 : 26 (g/mL), extraction 30 min, ullrasonic temperature 40 ℃. Under this condition, the total Iriterpenes extraction rate achieved at 3.06%. NKA-9 resin had the maximum adsorption amount for the purification total triterpenes and product purity reached 9.8%.
