Clinicopathological Significance of the Expression of Notch1 in Human Pancreatic Cancer

摘要 目的探讨胰腺导管腺癌(PDAC)中Notch1蛋白和mRNA表达水平及临床病理学意义。方法应用免疫组织化学方法检测57对配对的PDAC及癌旁组织石蜡标本Notch1蛋白表达水平,观察其与PDAC患者临床病理学参数的关系;再以实时定量聚合酶链反应(QRT-PCR)检测3株不同分化程度的胰腺癌细胞系(PANC-1、BxPC-3和CANPAN-2)中mRNA表达。结果免疫组化结果显示:Notch1在癌组织中表达高于配对的癌旁组织(t=2.459,P=0.020)。Notch1的表达与肿瘤分化程度呈负相关(χ2=7.155,P=0.028)。单因素及多因素分析发现Notch1是影响胰腺癌患者预后的一个独立因素(P值分别为0.015、0.026)。QRT-PCR结果显示:mRNA水平随细胞分化程度增高而逐渐减弱。结论在PDAC中Notch1表达明显上调,并与肿瘤分化程度相关。Notch1可以作为评估PDAC患者预后的一个独立指标。 Objective To explore the clinicopathological significance of the expression of notchl protein and mRNA levels in pancreatic ductal adenocarcinoma (PDAC). Methods The expression of nolchl protein in 57 pairs of para^nembedded PDAC specimens and adjacent non-cancerous pancreatic tissues were detected by immunohistocbemist^y. The relationship between the protein expression and clinicopatho]ogical characters was analyzed. Quantitative real-time polymerase chain reaction (QRT-PCR)were used to examine the mRNA expression of notch1 in 3 different differen- tiated pancreatic cancer ceils (PANC- 1, BxPC-3 & CAPAN-2 ). Results The expression of Notehl protein was higher in PDAC compared with that in paired non-cancerous pancreatic tissues ( t =2.459, P = 0.020 ).The expression of notchl protein had a negative correlation with tumor differentia- tion ( X2=7.155, P = 0.028 ). Univafiate and muhivariate analysis with clinicopathological characters revealed that the notch 1 expression was an inde- pendent prognostic indicator for PDAC patients ( P1 = 0.015 ; P2 = 0.026). Moreover, the mRNA expression of notch 1 was gradually reduced with the increase of differentiation degree of pancreatic cancer cell showed by QRT-PCR results. Conclusion Notchl is up-regulated in PDAC. It is cor- related with tumor differentiation. Notchl expression is an independent prognostic indicator for PDAC patients.
