摘要
目的:总结了醌氧化还原酶〔NAD(P)H:quinone oxidoreductase 1,NQO1〕在乳腺癌的发生发展中可能起到的作用及其与乳腺癌防治的联系。方法:应用PubMed、Springer Link及CNKI期刊全文数据库检索系统,以"乳腺癌(breast cancer)、NQO1、单核苷酸多态性(SNP)、雌激素(estrogen)"等为关键词,检索2000-2012年的相关文献,共检索到英文文献93篇,中文文献2篇。纳入标准:1)NQO1的生物学特征;2)NQO1的基因定位及其多态性;3)NQO1在乳腺癌中发生发展的作用及其表达;4)NQO1与乳腺癌的治疗及预后。符合分析的文献28篇。结果:NQO1作为人体内一种可被多种物质诱导的还原酶,在很多癌组织中表达异常,它具有多种功能,包括对苯醌等醌类物质产生的细胞毒性的解毒、氧化应激状态下稳定p53蛋白的表达、防止雌激素及其代谢产物对人类细胞的毒性和致癌作用等。NQO1频繁的基因多态性导致了个体中此酶活性的降低或缺乏,可能增加了乳腺癌的易感性。结论:NQO1对雌激素的作用使其在雌激素依赖性恶性肿瘤乳腺癌的发病及防治中有着重要意义,是一个强大的预后预测指标。
OBJECTIVE: To summarize the recent progress of NAD(P)H, quinone oxidoreductase 1 (NQO1) in de- velopment and prevention of breast cancer. METHODS: Related papers were searched with "breast cancer, NQO1, SNP, estrogen" as key words between 2000 and 2012 in PubMed, Springer Link and CNKI. Totally 93 English papers and 2 Chinese papers were obtained. A total of 28 papers were selected according to the standard as follows: Biological char- acteristics of NQO1. Assignment of NQO1 and its polymorphism. Function of NQO1 in development of breast cancer and its expression. Relation between NQO1 and the treatment and prognosis of breast cancer. RESULTS: NQO1 is a reduc- tase in human cells which can be induced by a variety of substances, it is expressed in multiped cancer tissues abnormally. It has various functions including the detoxification of quinone cytotoxicity, stable expression of p53 protein, prevention of the toxicity and carcinogenesis of estrogen and its metabolites to human cells. The polymorphism of NQO1 gene fre- quently leads to reduction even lack of this enzyme activity in the individual, thus it may increase the risk of breast cancer. CONCLUSIONS: As breast cancer is an estrogen-dependent cancer, NQO1 plays an important role in its pathogenesis, prevention and treatment by affecting estrogen. Consequently NQOI is a powerful prognostic indicator.
