东西方型幽门螺杆菌对Wnt/β-连环蛋白信号通路的影响 被引量:2

Effects of East Asian type and Western type Helicobacter pylori on Wnt/β-Catenin signaling pathway

摘要 目的探讨Wnt/β-连环蛋白(β-Catenin)信号通路在幽门螺杆菌(Hp)相关性胃癌发病机制中的可能作用。方法分别用东亚型和西方型Hp与人胃上皮细胞株(GES-1)共培养,Western blot检测细胞核内β-Catenin蛋白表达,荧光定量聚合酶链反应(RT-PCR)检测Wnt/β-Catenin信号通路下游c-myc和细胞周期蛋白D1mRNA表达,细胞一步法增殖检测试剂盒(MTS)检测细胞的增殖。结果东西方型Hp均引起了β-Catenin蛋白核内表达增加(P<0.01),但是东西方型菌株之间β-Catenin核内表达的增加没有统计学差异(P>0.05)。东西方型Hp处理细胞48h后,均引起了c-myc和细胞周期蛋白D1基因mRNA表达上调(P<0.05和P<0.01);但是东亚型组与西方型组间无统计学差异(P>0.05)。MTS检测显示,东西方型Hp处理细胞36h后,均引起了细胞增殖呈时间依赖的增加(P<0.01);并且在48h时,东亚型组促细胞增殖作用高于西方型组(P<0.05)。结论东亚型和西方型Hp均可激活Wnt/β-Catenin信号通路,促进胃癌的形成;但其作用在两种菌株之间无统计学差异。 Objective To investigate the possible role of Wnt/β-Catenin signaling pathway in Helicobacter pylori (Hp) infection-associated gastric cancer. Methods The gastric epithelial cells (GES-1) were co-cultured with two types of Hp[East Asian type(group E) and Western type(group W)]. The expression of β-Catenin protein in nucleus was detected by Western blot. The rnRNA levels of Wnt/β-Catenin signaling downstream c-myc and cyclin D1 were detected by fluorescent quantitative PCR. The proliferative potential of GES-1 cells was estimated by MTS method. Results The expression of β-Catenin in nucleus was increased in both groups (P〈0. 01 ), which was not significantly different between two groups(P〉0. 05). So did the expressions of c-myc and cyclin D1 mRNA after GES-1 cells were treated with two types of Hp for 48 hours(P〈0. 05 and P〈0. 01). MTS test showed that the cell proliferation was increased time-dependently in both groups after GES-1 cells were treated with two types of Hp for 36 hours(P〈0. 01) ,which was more in group E than that in group W after GES-1 cells were treated with two types of Hp for 48 hours(P〈0. 05). Conclusion East Asian type and Western type Hp could activate Wnt/β-Catenin signaling pathway, which may contribute to the development of gastric cancer. The effect is not significantly different between the two types of Hp.
