摘要
采集校园内腐质土壤,利用刚果红羧甲基纤维素钠初筛培养基、干稻草粉产酶培养基,通过水解圈与菌落直径的比值、滤纸酶活筛选及微生物鉴定得到高效降解植物纤维素的霉菌。结果表明,在刚果红初筛培养基上筛选出一株高产纤维素酶霉菌,其产滤纸酶活达到247.48U/mL。通过菌落形态、镜下形态及转化感受态细胞DH5α,鉴定出此霉菌为枝状枝孢霉(Cladosporium cladosporioides),具有较高的降解天然纤维素的能力,是一株有研究开发潜力的纤维素酶生产菌株。
This study focused on screening high efficiency cellulose decomposing fungus from humus in campus.The fungus were firstly screened through the ratio of hydrolytic circle and colony diameter which growing in Congo red plate, and then cultured in liquid medium with dry straw powder as carbohydrate supply for filter paper activity (FPA).The fungus were identified by microbiology at last.Results showed that the fungus was identified as Cladosporium cladosporioides by colony morphology,microscopic morphology and transformed cells DHSa.The filter paper activity of it was 247.48 U/mL.It would be a potential cellulose degrading fungus for research.
