摘要 目的探讨人体外周血冻存标本存放时间对提取DNA质量的影响。方法使用外周血DNA提取试剂盒FlexiGene DNA Kit(250),对-80℃冻存1a、2a、3a和4a的外周血标本进行DNA提取,NanoDrop-2000分光光度仪检测DNA浓度和OD260/OD280比值,1%琼脂糖进行电泳检测,采用单因素方差分析比较DNA质量。结果冻存1a、2a和3a的外周血DNA浓度之间无差异(P〉0.05),冻存4a的外周血提取DNA浓度明显低于冻存1-3aDNA的浓度(P〈0.05)。冻存1a的外周血DNA纯度明显高于冻存2a及以上的样品(P〈0.05)。结论外周血标本-80℃冻存时间超过3a对提取DNA的质量有明显的影响。 Objective To explore the impact for quality of DNA extraction from human peripheral blood samples by long time preservation. Methods eripheral blood was collected and stored at -80℃ for 1 year, 2 years, 3 years and 4 years. DNA extraction was performed using Flexi Gene DNA Kit (250). DNA concentrations and OD260/OD280 ratio were detected by NanoDrop 2000 spectrophotometer. Gel electrophoresis was performed by 1% agarose. One way ANOVA was applied to assess the DNA quality. Results The quality of the DNA extraction from human peripheral blood was similar on the blood samples with different preservation time of 1 to 3 years (P 〉 0.05). However, the quality of the DNA from samples with 4 years was significantly lower than those samples with 1 to 3 years (P 〈( 0.05). The purity of DNA extraction from the frozen samples with 1 years was significantly higher than those with 2 years and above peripheral blood (P 〈0. 0.05). Conclusion Stored at -80℃ for more than three years has apparently effect on the DNA quality extracted from human peripheral blood.
