摘要
目的探讨储存时间对石蜡包埋组织提取DNA质量的影响。方法采用试剂盒方法,分别提取10例保存1~3a的石蜡组织样品DNA,NanoDrop-2000分光光度仪检测DNA浓度和OD260/OD280比值,质量分数2%琼脂糖进行电泳检测,采用单因素方差分析比较DNA质量。结果琼脂糖凝胶电泳显示:石蜡包埋组织提取DNA降解明显。储存1~3a的石蜡包埋组织提取DNA的浓度有明显差异(P=0.04),储存时间越长,DNA浓度越低,但提取DNA的纯度无明显差异(P〉0.05)。结论石蜡包埋组织储存时间越长,提取DNA浓度越低,但纯度无明显差异。
Objective To explore the factors on quality and quantity of DNA extraction from human paraffin embedded tissues (PET) by store time. Methods DNA extraction was performed using DNA Kit from the PET stored for 1 year, 2 years and 3 years. DNA concentrations and OD260/OD280 ratio were determined by NanoDrop-2000 spectrophotometer. Gel electrophoresis was performed by 2 % agarose. One-way ANOVA was applied to assess the DNA quality. Results Agarose gel electrophoresis showed that DNA extraction from human PET degraded significantly. The concentration of the DNA extraction from human PET was significantly different in PET stored for 1, 2 and 3 years samples (P = 0.04), More longer the storage time, more lower the DNA concentration was. However, the purity of DNA was similar in PET stored for 1, 2 and 3 years. Conclusion More longer the storage time of PET, more lower the concentration of DNA extraction was, but no significant difference was observed in DNA purity.
