摘要
目的:探讨人参皂甙Rgl对体外与Ti微粒共培养的成骨细胞的影响及可能的干预作用。方法:体外培养新生大鼠成骨细胞,分别加入常规培养基、含胎牛血清的Ti微粒、Ti微粒+不同剂量人参皂甙Rgl的混合液培养。酶联免疫吸附法检测成骨细胞PGE-2、TNF-α、IL-1、IL-lra、IL-6的浓度;实时荧光定量PCR法检测COX-2、TNF-α mRNA表达;WESTRENBLOT法检测COX-2蛋白表达。结果:与Ti微粒共培养的成骨细胞给予人参皂甙Rgl干预后,细胞培养液中的PGE-2、TNF-α、IL-1、IL-6含量均明显低于单纯Ti微粒组,但高于正常对照组,成骨细胞的COX-2、TNF-α mRNA表达以及COX-2蛋白表达量也呈现同样的变化趋势。结论:人参皂甙Rgl能降低Ti微粒诱导的成骨细胞炎性细胞因子含量,对假体松动具有一定防治作用。
Objective:To explore the effect of ginsenosides Rgl with Ti particles on osteoblasts cultured in vitro and the pos-sible way of intervention. Methods:Osteoblasts from neonatal rats were cultured in vitro with conventional medium, fetal bovine serum with Ti particles or Ti particles along with different doses of Rgl. PGE-2, TNF-α, IL-1, IL-lra, IL-6, COX-2, TNF-α, mRNA, COX-2 protien in those three medium were detected by Enzyme linked immunosorbent assay, Re-al time fluorescence quantitative PCR method and WESTREN BLOT method respectively. Results:The level of COX-2, PGE-2, TNF-α, IL-1, and IL-6 in the Ti particles along with different doses of Rgl medium and COX-2, TNF-α mRNA COX-2 protien in cells of this group were higher than those of control group but lower than those of fetal bovine serum with Ti particles group. Conclusion:ginsenosides can reduce the levels of inflammatory cytokines produced by osteoblasts on in-duction with Ti particles and can prevent prosthesis loosening.
