摘要
观察疏肝降脂颗粒治疗非酒精性脂肪肝大鼠的治疗效果,并探讨该方的作用机制,从而为该方的临床应用提供实验依据。方法:将72只SD大鼠随机分为6组:空白组,模型组,多烯磷脂酰胆碱组,疏肝降脂颗粒低、中、高剂量组,每组12只。对除空白组外的其余5组大鼠均予高脂乳剂腹腔注射喂养8周,构建非酒精性脂肪肝大鼠模型。造模成功后,空白组予以标准饲料喂养,其余5组均继续以高脂乳剂灌胃喂养,并对各治疗组分别给予相应药物灌胃,空白组和模型组给予蒸馏水灌胃,连续12周后处死大鼠。实验过程中观察比较各组大鼠一般情况;实验结束前禁食12h,称取体质量,并计算肝指数;抽取空腹血分离血清后测定肝酶系、血脂,并观察肝脏病理变化。结果:与模型组相比,多烯磷脂酰胆碱组及疏肝降脂颗粒各剂量组均能较好的改善大鼠肝细胞脂肪变性(P<0.01,P<0.05),且疏肝降脂颗粒低、中、高剂量组之间的改善率比较呈现出量效正相关;疏肝降脂颗粒中、高剂量组与多烯磷脂酰胆碱组相比,肝脏病变明显减轻(P<0.05),以高剂量组效果最显著;在肝组织病理学方面,疏肝降脂颗粒各剂量组能减轻肝细胞损害程度,与多烯磷脂酰胆碱组比较,差异有统计学意义(P<0.05)。结论:疏肝降脂颗粒可以改善脂肪肝大鼠肝功能和血脂紊乱,减轻脂肪肝病变程度,从而达到治疗非酒精性脂肪肝的目的。
Objective :To observe the therapeutic eft^ct of liver- soothing and antihyperlipidemic granules in rats with non -alcohol- ie tarry liver disease (NAFLD) and investigate the action mechanism of this prescription, and to provide an experimental basis for clini- cal application of this prescription. Methods:Seventy- two Sprague -Dawley rats were randomly divided into blank group (n = 12 ) , model gruup ( n = 12 ), polyene phosphatidylcholine group ( n = 12 ) , and low - , middle - , and high - dose liver - soothing and antihy- perlipidenlie granule groups (n = 12 for each). All rats except those in blank group were intraperitoneally injected with high -fat cream for 8 weeks to establish a model of NAFI,D. After the model was successfully established,the blank group was fed with standard feed, and other groups continued to be fed with high - fat cream by gavage. The polyene phosphatidylchofine group amt liver - soothing and antihyperlipidemic granule groups received respective medicines by gavage,while the blank group and model group were given distilled water by garage. All rats were sacrificed alter 12 weeks of treatment. The general status was observed and compared between all groups during the experiment. Fasting was maintained for 12 h before the end of experiment,the body weight was determined,and the liver in- dex was calculated;fasting blood was drawn to separate serum and measure the levels of liver enzymes and blood lipids,and the patho- logical changes in the liver were evaluated. Results:Compared with the model group,the polyene phosphatidylcholine group and liver - soothing amt antihyperlipidemic granule groups had significantly reduced fatty degeneration of hepatocytes ( P 〈 O.
