多烯磷脂酰胆碱对NAFLD大鼠脂肪组织瘦素基因表达的影响 被引量:2

摘要 目的以分子生物学方法检测药物干预对NAFLD大鼠基因表达的影响。方法以多烯磷脂酰胆碱和蒸馏水灌喂NAFLD大鼠进行干预与对照实验,为期4周。提取大鼠脂肪组织的总RNA并逆转录成为cDNA,通过实时定量PCR方法检测两组大鼠的瘦素mRNA相对水平的变化。结果干预组大鼠脂肪组织瘦素mRNA的相对水平比对照组有所增加,但差异不显著(P>0.05)。结论多烯磷脂酰胆碱有助于增加脂肪组织瘦素mRNA水平和改善肝血脂质代谢。 Objective To detect the gene expression of NAFLD rats in the medical intervention using the methods of molecular biology. Methods The NAFLD mrs were intervened with polyene phosphatidylcholine or controlled with distilled water for 4 weeks. And then, the total RNA in adipose tissue was extracted and reversley translated into cDNA. The relative level of leptin mRNA was detected by the real-time quantitative polymerase chain reaction. Results The relative level of leptin mRNA in adipose tissue of intervenient group was higher than that of control group, but P 〉 0.005. Conclusion Polyene phosphafidylcholine is benefical to increase leptin mRNA level in adipose tissue and improve the adipose metabolism in liver and serum.
