β-D-葡聚糖和甘露聚糖对于人类健康有许多有益的作用,酵母细胞壁是其重要来源。啤酒废酵母泥是酿酒酵母中RNA提取的主要副产物。本文研究出一种以废酵母浆为材料分离β-D-葡聚糖和甘露聚糖的新方法。经过超声波法和高温高压处理后,β-D-葡聚糖是先用碱萃取后用蛋白酶酶解,收率和纯度分别为16.73%和76.82%。甘露聚糖是通过透析制备,最终产量和纯度分别为9.79%和64.39%。
The polysaccharides β-D-glucans and mannans show numerous beneficial effects for the health of humans and animals and the cell wall of Saccharomyces cerevisiae is an important source of β-D-glucans and mannans. Spent yeast slurry is the major by-product of the process of extracting RNA from Saccharomyces cerevisiae. We have developed a new method to isolate β-D-glucans and mannans with the material of spent yeast slurry. After sonication and high temperature and high pressure treatment, β-D-glucans were extracted firstly with alkali, then with protease, and then dried, with a yield of 16.73% and a purity of 76.82%. Mannans were prepared through the process of concentration, dialysis, precipitation and drying, with a finally yield and purity of 9.79%and 64.39%, respectively.
