摘要
目的基于"肺主通调水道"理论观察中药复方泽漆化痰方对烟熏合脂多糖(LPS)气道内滴入诱导的慢性阻塞性肺疾病(COPD)大鼠模型气道黏液高分泌的影响。方法 48只大鼠随机分为空白组、模型组、阳性药物沐舒坦对照组、泽漆化痰低剂量组、泽漆化痰中剂量组、泽漆化痰高剂量组,每组8只。于第30日处死大鼠,采集标本。采用HE染色和阿辛兰-过碘酸雪夫染色(AB-PAS)的组织学检查;免疫组化检测气管和肺内气道的MUC5AC、AQP5。结果与正常组比较,模型组大鼠气管黏膜上皮黏蛋白、MUC5AC蛋白表达升高,AQP5蛋白表达降低(P<0.05)。与模型组比较,泽漆化痰高剂量组黏蛋白、MUC5AC蛋白水平降低,AQP5蛋白水平升高(P<0.05)。与沐舒坦对照组比较,泽漆化痰高剂量组MUC5AC蛋白水平降低(P<0.05)。各剂量组对模型大鼠肺内气道的总黏蛋白、MUC5AC、AQP5水平的影响不明显。结论中药复方泽漆化痰方较高剂量具有抑制气道黏液高分泌的作用,且影响部位以大气道为主。
Objective:To study the influence of Zeqihuatan Decoction on airway mucus hypersecretion in rats based on the theory of lung governing regulation of water passages.Methods:48 wistar rats were randomly divided into blank group,model group,control group,asthmatic group,Zeqihuatan Decoction low dose group,Zeqihuatan Decoction middle dose group and Zeqihuatan Decoction high dose group,with 8 rats for each group.In thirtieth day the rats were killed to be collected as specimens.Histological examinations were performed by HE staining and AB-PAS staining; Protein MUC5AC and AQP5 were measured by immunohistochemistry.Results:Compared with normal group,tracheal epithelial mucin and MUCSAC protein expression increased and AQP5 protein expression decreased in model group(P< 0.05).Compared with model group,MUC5AC protein levels decreased and AQP5 protein levels increased in Zeqihuatan Decoction high dose group (P<0.05).Compared with control group,MUC5AC protein levels in Zeqihuatan Decoction high dose group decreased (P< 0.05).There was no statistical significance among pulmonary airways of different groups on total mucin,MUCSAC and AQP5 protein levels.Conclusion:This study shows that higher dose Zeqihuatan Decoction of Chinese herbal compound can inhibit airway mucus hypersecretion with large airway being the main point of action.
