人脑胶质瘤中PRDX-1、EGFR、PI3K、PTEN的表达及相关性 被引量:5

Expression of PRDX-1,EGFR,PI3K and PTEN in human glioma tissue and their correlation

摘要 目的探讨PRDX-1、EGFR、P13K和PTEN在不同恶性程度人脑胶质瘤中的表达及意义。方法用免疫组化sP法检测PRDX-1、EGFR、P13K及PTEN在156例人脑胶质瘤手术切除标本中的表达,Image—ProPlus6.0(IPP6.0)图像分析软件检测阳性部位平均吸光度值(A)进行定量分析,单因素方差分析及Dunnett’s T3检验比较组间差异,Pearson相关分析检验PRDX-1、EGFR、P13K及PTEN表达水平间的相关性。结果PRDX-1、EGFR及P13K的表达水平在I-Ⅳ级胶质瘤中差异显著(P〈0.05),且随胶质瘤恶性程度增加呈升高趋势;PTEN的表达水平在I~Ⅳ级胶质瘤中差异显著(P〈0.05),且随胶质瘤恶性程度增加呈降低趋势。PRDX-1与PTEN、PTEN与P13K在胶质瘤中的表达水平均呈显著负相关(r=-0.407、r=-0.429,P〈0.05);P13K与EGFR在胶质瘤中的表达水平呈显著正相关(r=0.293,P〈0.05)。结论PRDX-1、EGFR、P13K表达水平随胶质瘤等级上升而增高,PTEN表达水平则随胶质瘤等级上升而下降。上述蛋白可能通过PRDX-1/PTEN/EGFR/P13K途径在人脑胶质瘤的恶性演变中发挥重要作用。 Objective To study the expression of PRDX-1, EGFR, PI3K and PTEN in human glioma tissue and to seek for their possible relationship. Methods The expression of PRDX-1, EGFR, PI3K and PTEN in 156 case of glioma was examined with immunohistoehemical technique (SP), absorbanse (A) of the positive tissue was measured by Image-Pro Plus6. 0 for quantization, the statistical significances among groups were detected by One-Way ANOVA (Dunnett' s T3 ) and the relativities among the expression of PRDX-1, EGFR, P13K and PTEN were investigated by Pearson analysis. Results The expression of PRDX-1, EGFR and PI3K in glioma tissue showed significant differences among each grading ( P 〈 0.05 ), and increased with the degree of malignancy ; the expression of PTEN in glioma tissue showed significant differences between each grading ( P 〈 0. 05 ) , and decreased with the degree of malignancy. There were statistically significant correlation between the expression of PTEN and PRDX-1, PTEN and PI3K ( r = - 0. 407, r = - 0. 429, P 〈 0. 05 ) , and there was also a statistically significant correlation between the expression of EGFR and PI3K ( r = 0. 293, P 〈 0.05 ). Conclusions The expression of PRDX-1, EGFR and PI3K is increased with the ascending of pathological grading in glioma, and the expression of PTEN is declined with the ascending of pathological grading in glioma tissue. The PRDX-1/PFEN/ EGFR/PI3K pathway might be closely related to the occurrence and development of glioma.
