磷酸化mTOR及其下游分子4EBP1和S6K1在浸润性乳腺癌细胞中的定位表达研究 被引量:6

摘要 目的研究磷酸化mTOR及其下游分子4EBP1和S6K1在浸润性乳腺癌细胞中的表达部位。方法随机选取于2005年3月—2009年9月就诊于新疆医科大学附属肿瘤医院的285例乳腺癌患者,挑选乳腺癌组织及癌旁组织蜡块,重新切片,使用免疫组化方法检测磷酸化mTOR及其下游分子4EBP1和S6K1在细胞中的表达部位。结果在285例癌组织切片中,p-mTOR、p-4EBP1和p-S6K1的染色阳性例数分别为279例、161例和226例;在285例癌旁组织切片中,p-mTOR、p-4EBP1和p-S6K1的染色阳性例数分别为239例、81例和136例。p-mTOR在癌组织中表达定位于细胞核有1例,细胞浆有270例,核浆共表达有8例,在癌旁组织中表达定位于细胞核有0例,细胞浆有234例,核浆共表达有5例,癌组织与癌旁组织差异未见明显统计学意义(P=0.554);p-4EBP1在癌组织中表达定位于细胞核有3例,细胞浆有45例,核浆共表达有113例,在癌旁组织中表达定位于细胞核有1例,细胞浆有6例,核浆共表达有74例,癌组织与癌旁组织差异有统计学意义(P=0.001);pS6K1在癌组织中表达定位于细胞核有3例,细胞浆有28例,核浆共表达有195例,在癌旁组织中表达定位于细胞核有6例,细胞浆有0例,核浆共表达有130例,癌组织与癌旁组织差异有统计学意义(P=0.000)。结论mTOR信号通路中磷酸化4EBP1和S6K1在浸润性乳腺癌细胞浆中表达增高与乳腺癌相关。 Objective To investigate the location of phosphorelated mTOR and its downstream molecular 4EBP1 and S6K1 in invasive breast cancer cell.Methods 285 cases of breast cancer patients were randomly selected from March,2005 to September,2009 in the affiliated Tumor Hospital of Xinjiang Medical Uni-versity.The stored paraffin blocks of cancer and pericarcinomatous tissue were selected and recut into slices.Then P-mTOR, p-4EBP1 and p-S6K1 were detected by immunohistochemistry.Result The positive staining cases of p-mTOR,p-4EBP1 and p-S6K1 in carcinomaous tissue were 279,161 and 226,in pericarcinomatous tissue were 239,81 and 136.The cases of p-mTOR expressed in nuclear,cytoplasm,or co-expressed in nuclear and cytoplasm was 1,270 and 8 in cancerous tissue,0,234 and 5 in pericarcinoma-tous tissue,without statistically significance (P =0.554);The cases of p-4EBP1 were 3,45 and 113 in cancerous tissue,1,6 and 74 in pericarcinomatous tissue,with statistically significance (P =0.001);The cases of p-S6K1 were 3,28 and 195 in cancerous tissue,6,0 and 130 in pericarcinomatous tissue,with statistically significance (P =0.000).Conclusion Higher expression of cytoplasmic p-4EBP1 and p-S6K1 in invasive breast cancer cell may indicate the progression of breast cancer.
