hUCMSCs并西格列汀对糖尿病大鼠肝脏蛋白激酶B表达影响 被引量:1

THE EFFECT OF hUCMSCs COMBINED WITH SITAGLIPTIN ON EXPRESSION OF PROTEIN KINASE B IN LIVER OF RATS WITH DIABETES

摘要 目的 探讨静脉注射人脐带间充质干细胞(h U C MSCs)联合西格列汀治疗对链脲佐菌素(S T Z)诱导的糖尿病大鼠肝脏组织蛋白激酶B(AKT)表达的影响。方法 将32只大鼠随机分为正常对照组(8只)及糖尿病组(24只),糖尿病组采用高糖高脂饮食及腹腔注射小剂量STZ(30 mg/kg)方法制备糖尿病大鼠模型;成模后的糖尿病大鼠随机分为糖尿病对照组、hUCMSCs组及hUCMSCs联合西格列汀组(联合组),hUCMSCs组分别在成模后第1、5周行大鼠尾静脉注射hUCMSCs治疗;联合组成模后即开始给予大鼠西格列汀10 mg/(kg#d)灌胃治疗10周,第1、5周给予尾静脉注射h UC MS Cs治疗;糖尿病对照组不予治疗。造模成功后10周取大鼠肝脏组织,行免疫组化染色,比较各组大鼠肝脏组织AKT表达。结果 联合组及hUCMSCs组肝脏组织AKT表达均明显高于糖尿病对照组,低于正常对照组,差异有显著性(Hc=25.085,q=3 2.11~9.275,P〈0.05),联合组高于h UC MSCs组(q=5.967,P〈0 0.5)。结论 hUCMSCs治疗能改善因糖尿病引起的肝脏组织AKT表达降低,hUCMSCs与西格列汀联合能加强这种作用。 Objective To explore the influence of intravenous hUCMSCs combined with sitagliptin on expression of protein kinase B(PKB)in liver tissue of rats with diabetes induced by streptozotocin(STZ). Methods Thirty-two rats were divided into normal control group(eight rats)and diabetes group(24rats)in random.A diabetic model was created in diabetes group by high-glucose and high-fat diet and intraperitoneal injection of a small dose of STZ(30mg/kg).Upon completion of the model, the rats with diabetes were evenly randomized to diabetes-control group,hUCMSCs group and combined group(hUCMSCs+sitagliptin).The rats in hUCMSCs group were given caudal-vein injection of hUCMSCs on weeks 1and 5after creation of the model; those in combined group received sitagliptin 10mg/(kg·d)-starting on the day of model created-for 10weeks,and caudal-vein injection of hUCMSCs was offered on weeks 1and 5;no treatment was done in diabetes-control group.The rat livers were taken on week 10and immunohistochemical staining done.The expressions of PKB in liver of each group were compared. Results The expression of PKB in combined group and hUCMSCs group was higher than diabetes-control group,lower than normal control group,the differences were significant(Hc =25.085,q=3.211-9.275,P0.05),and that in combined group was higher than hUCMSCs group(q=5.967,P0.05). Conclusion The treatment with hUCMSCs can improve the reduced expression of PKB due to diabetes,and combined hUCMSCs with sitagliptin strengthens this action.
