摘要
利用变频微波萃取仪,辅助混合有机溶剂提取雨生红球藻中的虾青素,研究了单因素即提取温度、液料比与时间对虾青素提取率的影响,通过响应面试验优化提取参数,且检测了参数之间相互作用关系。利用统计分析和优化,结果显示,微波对虾青素作用的相对大小依次为时间>温度>液料比。通过计算回归方程优化得出液料比、提取温度和提取时间最佳萃取条件分别为200:1、45℃和20 min,在此条件下,虾青素提取率为36.88%。研究显示变频微波辅助混合有机溶剂萃取可快速提高雨生红球藻中虾青素提取率。
A combination of variable frequency microwave-assisted extraction was applied in identification of astaxanthin from Haematococcus pluvialis. The effects of experimental conditions on microwave- assisted extraction efficiency were studied, such as radio of solution to material, temperature, extraction time. Response surface methodology (RSM) was used for modeling and optimizing the process, and to gain a better understanding of the process performance. By RSM, the effects of radio of solution to material, temperature and time on the extraction efficiency were calculated and the interactions were determined. After statistical analysis and process optimization, the results showed that the effect of the extraction conditions was temperature〉time〉radio of solution to material, and the optimum conditions were: radio of material to solution 200:1, temperature 45 % and time 20 min, the assignment of astaxanthin was 36.88%. This study showed that the variable frequency microwave-assisted extraction can quickly enhance the extraction efficiency of astaxanthin from Haematococcus pluvialis.
