摘要
目的研究正源方(西洋参、当归、南沙参、仙鹤草、重楼)对60Co-γ射线辐射致小鼠造血和生殖系统损伤的保护作用。方法将小鼠随机分为正常组、模型组、皂矾丸组、正源方高、中、低剂量组。除正常组外其余组小鼠均接受5.5 Gy60Co-γ射线全身照射。观察各组小鼠白细胞、血小板计数和体质量的变化,实验结束后测定心、肝、脾、肺、肾、睾丸和精囊前列腺等脏器指数,在显微镜下观察脾脏和睾丸的病理变化。结果与模型组比较,各给药组小鼠的外周血白细胞、血小板计数及脾脏指数均显著上升(P<0.05或P<0.01),以正源方低剂量组上升最明显,其中血小板提升效果优于阳性药复方皂矾丸(P<0.01);正源方各给药组辐射小鼠的睾丸和精囊前列腺指数均有显著提高(P<0.01或P<0.05),以低剂量组最明显,而阳性对照药无该作用(P>0.05),脾脏和睾丸组织切片显示正源方低剂量组对辐射破坏的修复作用最好。结论正源方能够促进60Co-γ射线照射后小鼠外周血中白细胞、血小板的恢复,脾脏中造血细胞的再生以及睾丸中生殖细胞的增殖,对小鼠的造血和生殖系统损伤有一定的保护作用。
AIM To study the protective effect on hematopoietic and reproductive injuries of mice caused by 60-Co-γ ray irradiation of Zhengyuan Prescription (Panacis quinquefolii Radix, Angelicae sinensis Radix, Adenopho- rae Radix, Agrimoniae Herba, Paridis Rhizoma). METHODS Mice were randomly divided into control group, model group, Zaofan Pills group, and three Zhengyuan Prescription groups with high, medium and low dose, re- spectively. Except the control group, the other mice were exposed to whole-body irradiation of 5.5 Gy 60-Co-γ ray. White blood cells (WBC), platelets (PLT) and body weight were recorded after irradiation. The organ indexes of heart, liver, spleen, lung, kidney, testis and seminal vesicle prostate were measured and the pathological changes in spleen and testis were observed under the microscope. RESULTS Compared with the model group, the num- ber of peripheral WBC, PLT and spleen indexes of drug-treated group significantly increased (P 〈 0.05 or P 〈 0.01 ) , especially in the group with low dose of Zhengyuan Prescription. Importantly, low does of Zhengyuan Pre- scripition statitically improved the number of platelet than Zaofan Pills in mice (P 〈 0. 01 ). The indexes of the tes-tis and seminal vesicle prostate were highly improved in drug-treated group (P 〈0. 01 or P 〈0.05), and the inde- xes of the group with low dose of Zhengyuan Prescription improved the most, while Zaofan Pills group did not show similar results (P 〉 0.05 ) . Sections of spleen and testis tissue results showed that the group with low dose of Zhengyuan Prescription had a better curative effect on irradiation injury of mice. CONCLUSION Zhengyuan Prescription prevents the damages of hematopoietic organ and reproductive system in mice and promotes the recovery of peripheral WBC and PLT, the regeneration of hematopoietic cells in spleen, and the proliferation of germ cells in testis on mice irradiated by the 60-Co-γ ray.
