摘要
目的选择目前已有的辐射防护药物(包括化学药物、中药复方及细胞因子),研究联合用药的辐射防护活性,筛选最佳药物组合,为新的急性放射损伤治疗方案提供实验依据。方法试验分为模型对照组,E0703+rhG-CSF+新型四物合用组(组合Ⅰ),E0703+rhIL-11+刺白胶囊合用组(组合Ⅱ),E0703+rhG-CSF+新型四物合用组(组合Ⅲ),E0703+rhIL-11+刺白胶囊合用组(组合Ⅳ),523+rhG-CSF+新型四物合用组(组合Ⅴ),523+rhIL-11+刺白胶囊合用组(组合Ⅵ),523+rhG-CSF+刺白胶囊合用组(组合Ⅶ),523+rhIL-11+刺白胶囊合用组(组合Ⅷ),共9组药物组合。60Coγ射线7.5 Gy照射造模,照射前24 h按组分别给予化学药物预防,照后皮下注射细胞因子,并灌胃给予中药。照射前,照射后1、4、7、10、13、16、22 d检测小鼠外周血象。照射后第7天进行祖细胞集落培养实验。结果与模型对照组比较,除组合Ⅶ外,其余各组合对外周血液成分在不同时间点呈现不同程度的促进恢复作用;而对各造血祖细胞集落的生长,组合Ⅰ、Ⅲ的作用最为显著;其中,组合Ⅲ尚有促外周血白细胞、红细胞、血红蛋白值恢复的作用;组合Ⅴ也有加速外周血液成分恢复和促进粒系生长的作用。结论综合比较,组合Ⅲ对促进外周血液各成分恢复和造血祖细胞生长均有较好效果。
Objective To screen the best combination of radioprotectors by pretreating and curing cobalt-60 gamma irradiated mice with combined.Methods Synthetic compounds（E0703,523） were used as preventive agents,cytokine（rhG-CSF,rhIL-11） was used to cure early acute radiation damage,and tradiational Chinese medicine formulas（new si-wu capsules,ci-bai capsules） were used to reverse late radiation damage.Ninety mice were irradiated at a dose of 7.5 Gy using cobalt-60 gamma resources.Blood samples were collected from the tail end and the number of peripheral blood cells was counted on day 0,1,4,7,10,13,16 and 22 using a microcell counter.Hematopoietic progenitor colony-forming cell assays were used to assess the reconstitution of bone marrow after radiation-induced myelosuppression.Results Compared with control,white blood cells,red blood cells,hemoglobin and platelets were all increased in drug-treated groups,except for group Ⅶ.GroupⅠand group Ⅲ significantly increased the formation of hematopoietic progenitor colony-forming cell assays.Group Ⅲ also accelerated blood cell recovery.Group Ⅴ led to a significant increase in the formation of colony-forming unit-granulocyte macrophage and circulating blood cells.Conclusion Group Ⅲ is the best combination of radioprotectors.
