血链球菌细菌素对牙龈卟啉单胞菌生长曲线及细胞内钙离子浓度的影响 被引量:5

摘要 目的:研究血链球菌细菌素对牙龈卟啉单胞菌(Porphyromonas gingivalis P.g)生长曲线的影响。探讨血链球菌细菌素对牙龈卟啉单胞菌细胞内钙离子浓度的影响。方法:通过低温高速离心,超声破碎等方法提取血链素,使血链素作用P.g,于37℃厌氧培养:采用微量液体对倍稀释法测定血链素对P.g的最小抑菌浓度(MIC)。选用不同浓度的血链素作用P.g,分别于0、4、8、12、16、20、24、48h时测定各组细菌悬液在590nm波长处的A值,并绘制生长曲线。用Fluo-3/AM负载P.g细胞内钙离子,在激光共聚焦显微镜下观察Ca抖荧光强度变化。结果:血链素对P.g有显著抑制作用,其MIC为0.0625g/L。血链素作用后,P.g的生长曲线在8~16h发生显著改变,对数生长期被延迟了8h。血链素作用后,P.g细胞内钙离子浓度显著降低,与对照组相比差异具有统计学意义(P〈0.01)。结论:血链球菌细菌素对牙龈卟啉单胞菌有明显的抑制作用,在本实验范围内,随着血链素浓度的升高其抑制作用逐渐增强。血链球菌细菌素可导致牙龈卟啉单胞菌细胞膜钙离子通透性增强。 Objective: To study the effects of Streptococcus sanguis bacteriocins on the growth curve the intracellular free calcium concentration of Porphyromonas gingivalis. Methods.. We extracted Streptococcus sanguis bacterio- cins by low temperature high speed centrifugal and ultrasonic broken, made Streptococcus sanguis bacteriocins affect P. g at 37℃ anaerobic culture. Trace liquid times dilution method was developed for the determination of streptococcus sanguis bacteriocins of P. g minimum bacteriostatic concentration (MIC). We choose different concentrations of streptococcus sanguis bacteriocins affect P. g, respectively at 0, 4, 8, 12, 16, 20, 24 and 48 h to measure the determination of groups of bacteria suspension at 590 nm wavelength optical density (OD) value, and drew the growth eurve. We used Fluo--3/AM load P. g intraeellular calcium ions, and laser eonfocal microscope to observe Ca2+ flu- orescence intensity changes. Results: Streptococcus sanguis bacteriocins has a significant inhibitory effect of P. g,the MIC is 0. 0625 g/L. After the Streptococcus sanguis bacteriocins works, P. g intracellular calcium ion concentration decreased significantly. The growth curve of P. g has a significant change occurred in 8 - 16 h, logarithmic phase is delayed for eight hours. The intracellular calcium ion concentration of . P. g significantly reduced, statistically significant difference compared with the control group(P〈0.05). Conclusion: Streptococcus sanguis bacteriocins for P. g has obvious inhibition. Within the scope of this study, with the increase of streptococcus sanguis bacteriocins its inhibition enhanced gradually. Streptococcus sanguis bacteriocins can cause P. g cell membrane permeability enhaneed calcium bacterium.
