摘要
目的采用体内外试验方法研究蓼大青叶抗氧化活性。方法通过考察蓼大青叶体积分数95%甲醇提取液清除二苯代苦味酰基自由基(DPPH·)活性以及对H2O2诱导小鼠红细胞氧化溶血的影响,评价其体外抗氧化活性;通过研究蓼大青叶体积分数95%甲醇提取液对CCl4所致急性肝损伤小鼠血浆中总超氧化物歧化酶(T—SOD)活力、丙二醛(MDA)水平及肝匀浆液中谷胱甘肽过氧化物酶(GSH—Px)活力的影响,探讨蓼大青叶的体内抗氧化活性。结果蓼大青叶体积分数95%甲醇提取液质量浓度在25—1000mg·L-1。内可有效清除DPPH·,IC50 为103.4mg·L-1;蓼大青叶体积分数95%甲醇提取液质量浓度在25~100mg·L-1内具有H2O2诱导红细胞氧化溶血抑制作用;剂量范围在3~12g·kg-1内能减少小鼠血浆中MDA水平,增加T—SOD活性,提高肝匀浆液GSH—Px活性。结论蓼大青叶体积分数95%甲醇提取物具有一定的体内外抗氧化活性。
Objective To observe the antioxidant activity in vivo and in vitro of the Polygoni Tinctorii Foli- um. Methods The in vitro antioxidant activity of the Polygoni Tinctorii Folium was studied according to the scavenging rate of DPPH. and the influence on the oxygen hemolysis of red blood cells induced by H2 O2 with 95 % methanol extraction of the Polygoni Tinctorii Folium. The in vivo effect was observed by determi- ning the values of SOD, MAD in the plasma and GSH-Px in the liver homogenate of mice. Results The 95 % methanol extraction of the Polygoni Tinctorii Folium could significantly scavenge DPPH. with IC50 of 103.4 mg.L- 1in the range of 25- 1 000 mg. L-1, and inhibit the hemolysis of red blood cells induced by H2O2 between 25- 100 mg .L-1. It could also lower the level of MDA, increase the activity of T-SOD in mice plasma, and improve the vitality of GSH-Px in mice liver homogenate within the dosage of 3- 12 g. kg-1. Conclusions The 95 % methanol extraction of Polygoni Tinctorii Folium has obvious antioxidant activity in vivo and in vitro.
