摘要
目的:探讨Ilexgenin A诱导人宫颈癌HeLa细胞凋亡及其相关机制。方法:采用4',6-二脒基-2-苯基吲哚(DAPI)进行核染色观察细胞凋亡形态学变化,Apo-ONEHomogeneous均质蛋白酶-3/7检测试剂盒用于测量Caspase-3/7活性,通过测定宫颈癌细胞核提取物NF-κB p65与DNA结合活性来衡量NF-κB的激活水平。结果:DAPI荧光染色发现,IA作用24h后,细胞核浓缩染色质凝聚,凋亡小体出现;IA以剂量依赖的方式显著增加了Caspase-3/7的活性;IA对NF-κB的p65亚基的活化有显著的抑制作用,并且呈一定程度的剂量依赖性。结论:IA可明显诱导宫颈癌HeLa细胞凋亡,并呈浓度依赖性。
Objective: To study the Ilexgenin A-induced apoptosis in cervical cancer HeLa cells and Its mechanisms. Methods : DAPI was adopted for nucleus stain to observe the morphoiogy change during apoptosis ; Apo-ONE Homogene- ous protease-3/7 kit was used for measure the activity of Caspase-3/7 ; and the binding activity of nuclear extracts of cervi- cal cancer HeLa cells NF-KB p65 with DNA was determined to investigatethe activation level of NF-KB. Results: DAPI assay showed that Karyopyknosis and chromatin condensation occurred and apoptotic bodies appeared 24 h after Ilexgenin A(IA) induction. IA enhanced the activity of Caspase-3/7 at the manner of dose-dependence. IA assumed remarkable inhibitory effect on the activation of subunit p65 of NF-KB, and this inhibitory effect was found dose-independent to some extent. Conclusion: IA could remarkably induce the apoptosis in cervical cancer HeLa cells at the manner of concentration dependence.
