Expressions and significances of Xiap and Survivin in placental tissues of patients with preeclampsia

摘要 目的:检测Xiap和Survivin在子痫前期患者胎盘组织中的表达,研究滋养细胞凋亡与子痫前期的关系。方法:用免疫组化SP法和DNA原位末端标记TUNEL法分别测定30例轻度子痫前期、30例重度子痫前期和30例正常晚期妊娠胎盘组织中Xiap和Survivin蛋白的表达和细胞凋亡,用计算机图像分析系统定量分析。结果:Xiap和Survivin位于绒毛小叶的合体滋养细胞及细胞滋养细胞,在子痫前期的表达显著低于正常晚期妊娠(P<0.05),随病情加重表达降低;Xiap和Survivin的蛋白表达与细胞凋亡指数呈负相关,子痫前期Xiap和Survivin表达呈正相关(P<0.05)。结论:子痫前期患者胎盘组织中Xiap和Survivin的减少可能与子痫前期的发生有关。 Objective: To detect the expressions of Xiap and Survivin in placental tissues of patients with preeclampsia, in order to study the relationship between apoptosis of trophoblasts and preeclampsia. Methods: The expression levels of Xiap and Survivin and apop- totic rates in placental tissues of 30 patients with mild preeclampsia, 30 patients with severe preeclampsia, and 30 women of normal late pregnancy were detected by immunohistocbemical SP method and TUNEL method; then the results were analyzed quantificationally by com- puter image analysis technique. Results: Xiap and Survivin were mostly found in syncytiotrophoblast and cytotrophoblast. The expressions of Xiap and Survivin in preeclampsia group were statistically significantly lower than those in normal late pregnancy group (P 〈 0.05 ), which decreased with aggravation of the disease. The expressions of Xiap protein and Survivin protein were negatively correlated with apoptosis in- dex. There was a positive correlation between Xiap expression and Survivin expression in preeclampsia group ( P 〈 0.05 ) . Conclusion: Down - regulations of Xiap and Survivin in placental tissues of patients with preeclampsia may be correlated with the onset of preeclampsia.
