摘要
目的分析葡萄糖神经酰胺合成酶(glucosylceramide synthase,GCS)和P-糖蛋白(P glycoprotein,P-gp)在胃癌组织中的表达及相关性,及二者与临床病理特征的关系。方法采用免疫组化SP法检测132例胃癌组织及癌旁组织中GCS和P-gp蛋白的表达;应用实时荧光定量PCR技术检测胃癌组织中GCS和P-gp mRNA的表达。结果 (1)胃癌组织中GCS、P-gp阳性率均高于癌旁组织(P<0.01)。(2)胃癌组织中GCS与P-gp表达呈正相关(P<0.001)。(3)胃癌组织中GCS和P-gp mRNA表达呈正相关(P<0.01)。(4)胃癌中GCS阳性率与肿瘤浸润深度、淋巴结转移以及TNM分期有关(P<0.05);P-gp阳性率与肿瘤大小、分化程度、浸润深度以及TNM分期有关(P<0.05)。结论胃癌组织中GCS不仅和P-gp与多药耐药有关,且可提示肿瘤的浸润、转移。
Purpose To explore the role of expression of glycosylceramide synthase (GCS) and P glycoprotein (P-gp) in multidrug re- sisitance of gastric carcinoma, and to analyze their clinicopathologic significance. Methods Immunohistochemical SP method was per- formed to investigate the GCS and P-gp expression in 132 cases of gastric carcinoma and their surrounding normal tissues. Real-time PCR was adopted to analyze the correlation of GCS and P-gp mRNA in gastric carcinoma. Results ( 1 ) The expression of GCS and P- gp was higher in the carcinoma than their surrounding normal tissues (P 〈 0. 01 ). (2) The positive correlation of GCS and P-gp ex- pression existed in the gastric carcinoma ( P 〈 0. 001 ). (3) The positive correlation of their mRNA level also existed in the carcinoma (P 〈 0.01 ). (4) The expression of GCS was positively correlated to infiltration, lymph node metastasis and TNM stages of gastric car- cinoma ( P 〈 0. 05), while that of P-gp was positively correlated to size, differentiation, infiltration and TNM stages of gastric carcino- ma (P 〈 0. 05 ). Conclusions The expression of GCS in gastric carcinoma is not only closely related to P-gp and muhidrug resistance but also related to infiltration and metastasis.
