摘要
目的:探讨壮肾排毒方抗肾纤维化的作用机制,为临床应用提供实验依据。方法:48只雄性Wistar大鼠随机分为正常组、模型组、壮肾排毒方组、福辛普利组各12只。除正常组外其余各组均行5/6肾切除手术制作慢性肾衰竭(CRF)动物模型。于造模第2次手术后1周开始干预,干预12周后取血清及肾组织标本,全自动生化分析仪检测血清尿素氮(BUN)、血肌酐(Scr)。免疫组织化学方法检测肾组织血肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)的表达。结果:模型组、壮肾排毒方组、福辛普利组大鼠血清BUN、Scr肾脏组织TNF-α、TGF-β1与正常组比较有显著性差异(P<0.01);模型组血清BUN、Scr及肾脏组织TNF-α、TGF-β1与壮肾排毒方组、福辛普利组比较有显著性差异(P<0.05,P<0.01)。结论:壮肾排毒方是治疗CRF的有效方剂,其机理可能与抑制大鼠肾组织内TNF-α及TGF-β1的表达有关,可抑制参与肾纤维化的细胞增殖和转分化,延缓肾纤维化的进展,从而达到治疗CRF的目的。
Objective: To explore the mechanism of Zhuangshen Paidu Fang on chronic renal failure ( CRF ) and renal fibrosis and provide the experimental evidences for clinical application.Methods: 48 Wistar male rats were divided into fourgroups randomly, namely normal control group ( shamoperation group, 12rats ) , model group ( 12 rats ) , Zhuangshen Paidu group ( 12rats ) and Fosinopril group ( 12 cases ) .Except the rats in shamoperation group, CRF models were prepared with 5/6nephrectomy in the rest groups.Intervention started lw after the second operation of modeling. 12w after intervention, serum and kidney tissue specimen were collected.Automatic biochemistry analyzer was applied for the determination of BUN and Scr .Immunohistochemistry was for the test of the expression of TNF-α and TGF- β 1 in kidney tissue.Results: The significant difference was presented in serum BUN and Scr, TNF-α and TGF- β1 in kidney tissue in comparison of model, Zhuangshen Paidu group and Fosinopril group with shamoperation group ( P〈0.01 ) .Comparing with Zhuangshen Paidu group and Fosinopril group, the outcomes of serum BUN, serum Scr and TNF- α, TGF- β1 in kidney tissue in model group were different significantly (P〈0.01, P〈0.05 ) .Conclusion: Zhuangshen Paidu Fang is the effective formula for CRF, which is probably relevant with the inhibition of cellular factor expression, like TNF- α, TGF- β1 in kidney tissue in rats to prohibit the proliferation and transdifferentiation of ceils that are involved in renal fibrosis; thus, to aim at the treatment and delay the progress of CRF.
