摘要
目的:研究人体病原真菌白念珠菌CaPsy2蛋白的功能,探讨它在DNA损伤检查点修复过程中所起的作用。方法:采用同源重组的方法构建CaPSY2双等位基因缺失株,并利用倍比稀释法探讨CaPSY2缺失株在含有遗传毒性试剂平板上的表型。结果:PCR检测结果显示CaPSY2双等位基因缺失株构建成功,而表型结果显示CaPsy2缺失会导致缺失株对遗传毒性试剂敏感,且这种敏感性表型可以被外源的CaPSY2基因互补。结论:CaPsy2蛋白参与DNA损伤修复过程,并且可能作为CaPph3的调节亚基而发挥作用。
Objective:To study the gene function of CaPSY2 in DNA damage checkpoint in Candida albicans. Methods:Ho-mologous recombination was used to construct the null mutant of CaPSY2 and phenotype screening was used to explore the function of CaPsy2. Results: Homozygous null mutant for CaPSY2 was constructed successfully and showed hypersensitive to genotoxicity which can be rescued by exogenous CaPsy2. Conclusion: CaPsy2 plays a role in DNA damage checkpoint path-way in Candida albicans.
