5-Fu concentration in submandibular lymphatic nodes and tongue after continuous infusion via lingual artery or femoral vein

摘要 目的:比较舌动脉及股静脉持续灌注5-氟尿嘧啶(5-Fu)后,不同时间点舌及区域淋巴结组织、血浆中的药物浓度。方法:16只健康家犬称重后随机分为A组(舌动脉给药)和B组(股静脉给药),2组均持续24 h灌注5-Fu。在6、12、18、24 h分别切取灌注侧舌体及下颌下淋巴结;在不同时间点采集对侧股静脉血浆样品。反向高效液相色谱法检测(r-HPLC)组织样品及血浆中的5-Fu含量。结果:在各时间点,A组置泵侧舌体及下颌下淋巴结内药物浓度明显高于B组(P<0.01)。给药2 h后B组血浆浓度均高于A组(P<0.05)。结论:5-Fu舌动脉持续灌注后,同侧下颌下淋巴结及舌组织中浓度明显高于全身灌注,血浆药物浓度低于全身灌注。 Objective: To compare the concentrations in submandibular lymphatic nodes and tongue after continuously infusion of 5- Fu via lingual artery or femoral vein. Methods: 16 dogs were randomly divided into to 2 groups(n =8). Dogs in group A and B re- ceived lingual artery and femoral vein 24 h continuous infusion respectively. The ipsilateral submandibular lymphatic nodes and tongue tissue were harvested, blood plasma samples were collected at different time points. 5-Fu concentration was determined by re- verse- High- Performance Liquid Chromatography ( r- HPLC ). Results: 5- Fu concentration in ipsilateral submandibular lymphatic nodes and tongue of group A was significantly higher than that of group B(P 〈0.01 ). Plasma 5-Fu level of group A was lower than that of group B after 2 h infusion(P 〈0.05). Conclusion: Continuous lingual artery infusion of 5-Fu may produce higher concentra- tion of the drug in ipsilateral submandibular lymphatic nodes and tongue than femoral vein infusion, and lower in plasma.
