摘要
目的通过观察金欣口服液(炙麻黄、苦杏仁、生石膏、黄芩等)对呼吸道合胞病毒(RSV)感染肺组织TOLL样受体4(TLR4)及转化生长因子β活化激酶(TAK1)表达的调控趋势,探讨金欣口服液抗病毒机制以及TLR4信号通路途径。方法 RSV滴鼻感染BALB/c小鼠,金欣口服液给药进行干预,并于首次滴鼻后24 h取各组小鼠肺组织,Western Blot法检测小鼠肺组织TLR4、TAK1蛋白表达。结果 RSV感染BALB/c小鼠后24h,TLR4﹑TAK1蛋白表达明显升高(P<0.01),而金欣口服液组较RSV感染组TLR4﹑TAK1表达明显下调(P<0.01);金欣组蛋白表达量最低,与利巴韦林组相比,P<0.01,利巴韦林组与RSV感染组相比P<0.01。结论金欣口服液能抑制TLR4信号转导通路中TLR4/TAK1蛋白的表达,TLR4的蛋白表达与TAK1的蛋白表达呈正相关,金欣口服液抗RSV作用机制中的TLR4信号通路途径可能是通过TAK1实现的。
ABSTRACT: AIM To study the effect of Jinxin Oral Liquid (Ephedrae Herba, Armeniacae Semen amarum, Gyp- sum fibrosum, Scutellariae Radix, etc. ) on the expression of Toll-like receptor4 (TLR4) and TAK1 of mice lung tissues infected with respiratory syncytial virus (RSV) and explore Jinxin Qral Liguid's anti-viral mechanism and TLR4 signal path. METHODS BALB/c mice were intranasally inoculated with RSV, and then treated with Jinxin Oral Liquid. After 24 hours of inoculated with RSV, the mice were killed and the lung samples were collected, and then the expression of TLR4 and TAK1 was measured by Western Blot. RESULTS The results demonstrated that after being infected twenty-four hours, the expression of TLR4 and TAK1 were up-regulated significantly (P 〈 0. 01 ), but in oral liquid group were down-regulated compared with the RSV infected group ( P 〈 0. 01 ). The level of protein expression in Jinxin group was the lowest, compared with that in Ribavirin group (P 〈 0. 01 ), and the Ribavirin group compared with the RSV infected group (P 〈 0. 01 ). CONCLUSION Jinxin Oral Liquid can inhibit the expression of TLR4/TAK1 in signal transducing pathway of TLR4 in a positive correlation between TLR4 and TAK1, the antiviral mechanism of TLR4' signal transducing pathway may be related to TAK1.
