摘要
目的探讨人心房利钠肽(hANP)对大鼠机械通气性肺损伤(VILI)时炎症因子和NF-κB p65表达的影响。方法采用高气道压机械通气模式制备机械通气性肺损伤模型。雄性SD大鼠30只随机分为空白对照组、机械通气肺损伤组(HV组)和人心房利钠肽组(HV+hANP组)。HV+hANP组于机械通气30min静脉注射hANP(0.1g/kg·min),机械通气4小时时处死大鼠,免疫蛋白印迹法测定肺组织NF-κB p65的表达,酶联免疫吸附法测定肺组织TNF-a和IL-lβ含量,计算肺组织湿/干重比(W/D)。结果与TV组相比,HV组、HV+hANP组肺组织NF-κB p65表达明显增加,肺组织TNF-a和IL-lβ含量明显上升(P<0.05),肺组织W/D明显升高(P<0.05);与HV组相比,HV+hANP组肺组织NF-κB p65表达明显抑制,肺组织TNF-a和IL-lβ含量明显降低(P<0.05),肺组织W/D降低(P<0.05)。结论人心房利钠肽通过抑制肺组织NF-κB p65表达,减轻肺部炎症反应,从而减轻大鼠机械通气所致肺损伤。
Objective To investigate the effects of human atrial natriuretic peptide (hANP) on the expression of nuclear factor-kappaB (NF-κB) p65 and content of TNF-a and IL-1β in the lungs induced by ventilator (VILI). Methods A rat model of lung injury induced by ventilation with high peak inspiratory pressure. Thirty male SD rat weighing 200- 250g were randomly divided into 3 groups(n= 10 each) .. group TV(traditional tidal volume VT= 8mL/kg), group HV (high tidal volume VT=40 mL/kg) and group HV+ hANP(treated with hANP 0. 1g/kg, rain after 30 min ventilation). The animals were sacrificed at 4h after mechanical ventilation. The lungs were immediately removed for determination of expression of NF-rB p65(Western blot ) , TNF-a and IL-I~ content(enzymelinked immuno- sorbentassy, ELISA ) and wet to dry weight ratio (W/D). Results Compared with group TV, the expression of NF-rB p65 in the cell nucleus the level of TNF-a and IL-1β and W/D in the lungs was significantly increased in group HV and group HV+ hANP compared with group HV (P〈0. 05). Conclusion hANP treatment can attenuate the changes in NF-~B p65 expression in the lung induced by ventilation, hANP may alleviate the inflammation of acute lung injury in rat.
