钙非依赖磷脂酶A_2参与非L型钙通道开放引起的心肌细胞缺氧复氧损伤 被引量:2

摘要 目的:探讨钙非依赖磷脂酶A2(iPLA2)是否参与胞外无钙缺氧复氧(H/R)损伤。方法:制作无钙H/R模型,观察不同H/R时间点iPLA2和超氧化物歧化酶(SOD)的表达。在无钙H/R条件下,利用iPLA2质粒转染心肌细胞株(H9C2)使iPLA2过表达,通过检测氧自由基指标SOD活性、丙二醛(MDA)浓度和炎症指标花生四烯酸(AA)分泌来观察细胞损伤情况;利用SiRNA干扰使iPLA2基因沉默,检测上述指标,观察细胞损伤情况。结果:无钙H45 min/R30 min时,iPLA2蛋白表达最高且SOD活性较正常组有显著性下降(P<0.05);在无钙H45min/R30min条件下,转染iPLA2的质粒后,可使iPLA2高表达,SOD活性降低,MDA、AA浓度升高(P<0.01);转染iPLA2的SiRNA进行干扰后,可使iPLA2低表达,提高SOD活性,降低MDA、AA浓度(P<0.01)。结论:iPLA2参与了无钙H/R损伤,高表达可加重损伤,低表达可减轻损伤。
