川穹嗪联合胚胎干细胞治疗大鼠肺纤维化的机制研究 被引量:10

摘要 目的:研究静脉注射胚胎干细胞(ESC)和川穹嗪(TMP)对肺纤维化大鼠的治疗作用。方法:气管内滴注博莱霉素8.5mg/kg制作雌性大鼠的肺纤维化模型。治疗组分别单独(联合)静脉注射大鼠ESC与川穹嗪,对照组注射生理盐水。记录大鼠的生存状态,测定大鼠肺组织的羟脯氨酸含量,肺脏病理学观察炎症状态、细胞凋亡情况以及相关蛋白的变化。结果:与对照组相比,接受ESC与TMP联合治疗后,肺纤维化模型大鼠的体质量增加,状态良好;肺羟脯氨酸含量明显降低;肺脏病理检查显示肺组织炎症程度降低,结构破坏减轻。流式细胞结果显示细胞凋亡指数降低,其核心蛋白MMP-2和MMP-9的表达明显下调。结论:静脉注射ESC与TMP可以减轻博莱霉素诱导的大鼠肺部炎症和肺纤维化。 Objective: To investigate the therapeutic effect of tetramethylpyrazine and embryonic stem cells in pulmonary fibrosis of rats. Methods: IntratracheaI instillation of the bleomycin 8.5 mg/kg to produce female rats model of pulmonary fibrosis. Treatment groups were injected with ESC or/and Chuanqiongqin, compared with the control group with saline. The survival state of the rats were recorded. Meanwhile, the content of hypdroxypro- line (Hyp) in lung tissue was assayed. Detection of the lung pathology, cell apoptosis and the activities of apoptosis related proteins were processed. Results: Compared with the control group, the group of ESC and TMP therapy had a good condition with the increasing of body weight; lung hydroxyproline content was significantly reduced; lung pathological examination showed a lower degree of lung tissue inflammation, with the mitigation of structural dam age. The apoptotic index was decreased by flow cytometry, and the expression of MMP-2 and MMP-9 were signifi cantly down regulated. Conclusion:ESC and TMP might be a therapeutic medicine for pulmonary fibrosis.
