摘要
目的建立同时测定酸枣叶中芦丁、槲皮素-3-O-β-D-葡萄糖苷、山奈酚-3-O-芸香糖苷和槲皮素-3-O-α-L-阿拉伯糖基(1→2)-α-L-鼠李糖苷含量的方法。方法采用HPLC法。色谱柱为Agilent C18柱(150mm×4.6mm,5μm),流动相为乙腈(A)-体积分数为1%的乙酸溶液(B)梯度洗脱,流速0.8mL·min^-1,检测波长255nm,柱温35℃。结果酸枣叶中4个指标成分芦丁、槲皮素-3-O-β-D-葡萄糖苷、山奈酚-3-D-芸香糖苷和槲皮素-3-O-α-L-阿拉伯糖基(1→2)-α-L-鼠李糖苷质量浓度分别在13.93-557.2mg·L^-1(r=0.9998)、1.030-41.20mg·L^-1(r=0.9999)、1.040-41.60mg·L^-1(r=0.9999)和7.420-296.8mg·L^-1(r=0.9999)内与峰面积呈良好的线性关系。加样回收率分别为97.4%、97.7%、98.1%和97.6%,RSD分别为2.2%、2.9%、3.1%和1.9%。结论该方法操作简便、快速、准确、重复性好,可作为酸枣叶的含量测定方法.为酸枣叶的质量摔制和开发利用提供实验基础。
Abstract: Objective To establish an HPLC method for the simultaneous determination of rutin,quercetin-3- O-β-D-glucopyranoside, kaempferol-3-O-α-L-rhamnopyranosyl ( 1 ±6 ) -β-D-glucopyranoside, quercetin-3-O- α-L-arabinopyranosyl (1→2) -α-L-rhanmopyranoside in leaves of Ziziphus jujuba var. spinosa. Methods An Agilent ODS C18 column ( 150 mm ×4. 6 mm,5 μm) was used with a gradient elution of acetonitrile-water containing 1% acetic acid. The flow rate was 0. 8 mL· min^-1 , the detection wavelength was set at 255 nm and the column temperature was 35 ℃. Results The method had a good linearity in the ranges of 13.93 - 557.2 mg· L ^-1 ( r = 0. 999 8 ) for rutin, 1. 030- 41.20 mg· L ^-1 ( r = 0. 999 9 ) for quercetin-3-O-β-D-glucopyranoside, 1. 040- 41.60 mg· L ^-1 ( r = 0. 999 9 ) for kaempferol-3-O-α-L-rhamnopyranosyl ( 1 →6 ) -β-D- glucopyranoside, 7. 420- 296.8 mg· L^ - 1 ( r = 0. 9999 ) for quercetin-3-O-α-L-arabinopyranosyl ( 1→2 ) -α-L- rhamnopyranoside,respectively. The average recoveries for those four standard substances were 97.4%, 97.7% ,98.1% and 97.6% ,with RSD of the recoveries of 2. 2% ,2. 9% ,3.1% and 1.9% ,respectively. Conclusions The method is simple, rapid, accurate and reproducible, and it can provide the basis for the quality control and exploitation of leaves of Ziziphus jujuba var. spinosa.
