瓜蒌皮注射液干预急性心肌梗塞药效与舒张血管机制的研究 被引量:15

Protective Effect and Underlying Mechanism of Trichosanthes Pericarpium Injection on Acute Myocardial Infarction Rat Model

摘要 目的:研究瓜蒌皮注射液干预急性心肌梗塞的作用效果,并探讨其机制。方法:结扎大鼠冠脉动脉,制备冠心病急性心肌梗死模型。丹参注射液为阳性对照药。大鼠腹腔注射给药。氯化三苯四氮唑染色(TTC),测定心肌梗死率;生化试剂盒检测血浆中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、谷草转氨酶(AST)、羟基丁酸脱氢酶(α-HBDH)和乳酸脱氢酶(LDH)5项心肌酶活性,确定心肌细胞坏死程度;生化试剂盒检测血浆中内皮素(ET)、一氧化氮(NO)含量及内皮型一氧化氮合酶(eNOS)的活性,明确瓜蒌皮注射液调节血管舒张的能力。结果:对急性心肌缺血大鼠,瓜蒌皮注射液可显著降低其心肌梗死率及血浆中5项心肌酶活性,提高eNOS活性及NO分泌量,降低ET含量,维护缺血心肌正常生理功能,作用效果与丹参注射液无显著性差异。结论:瓜蒌皮注射液可能通过抑制ET的合成、激活eNOS活性,提高NO分泌量,增强冠状动脉的扩张,增加冠脉流量;从而实现对大鼠急性心肌梗塞的保护作用。 Objective:Trichosanthes pericarpium injection results from the traditional Chinese medicine Trichosanthes pericarpi- urn. This medicine had been used to treat coronary heart disease patients for two thousand years in China. This study aims to eval- uate the pharmacodynamical effect in myocardial infarction protection and to explore the underlying mechanism of this protection. Methods:To make acute myocardial ischemia animal model, 1 hour after the last administration, the left anterior descending coronary branch were ligated in male Wistar rats received abdominal cavity administration for 7 days. Rats in different groups were giv- en Trichosanthes pericarpium injection,the radix salviae miltiorrhizae injection (the positive control drug),and 0.9% sodium chloride solution. The degree of myocardial infarction was evaluated by electrocardiograph (ECG). 6 h after of operation, the in- farction area and the infarction rate of myocardial tissue in every rat was checked after being stained with triphenylte trazoliam chloride (TrC). The myocardial injury, activity of creatine-kinase ( CK ), creatine-kinase - MB ( CKMB), lactate dehydrogenase (LDH) and aspartate- amino transferase(AST) in serum were detected in these rats of different groups. Moreover, in order to find out the mechanism by which Trichosanthes pericarpium injection to protect myocardial infarction, the serum contents of endo- thelin (ET), nitric oxide (NO) and endothelial nitric oxide synthase (eNOS) in these rats were measured. Results:Compared with that in the model group rats, the myocardial infarction rate of rats in the Trichosanthes group was decreased evidently( P 〈 0. 05 ), the serum activities of CK, CK- MB, LDH, AST and c^- HBDH were also obviously low (P 〈 0. 05 or P 〈 0. 01 ). The serum ET content was markedly reduced, but the NO level and the activity of eNOS in the serum were increased observably ( P 〈 0. 05 ).
