摘要
用猪精液抑制素 ( inhibin,IB)抗原主动免疫供体母牛 ,结合外源纯化 FSH超排处理 ,观察了抑制素主动免疫对牛超排效果的影响 ,并通过检测不同生理时期血液中相关生殖激素的水平 ,探讨了抑制素免疫影响牛超排效果的机制。选产后 50 d以上的 3~ 5岁荷斯坦母牛 1 2头和西杂青年母牛 1 2头 ,按其发情顺序随机分为试验组和对照组 ,每组 1 2头。试验组用完全福氏佐剂乳化的 IB抗原进行首免 ,间隔 2 0 d再用不完全福氏佐剂乳化的 IB抗原加免 1次 ,加免后 6 d开始采用纯化 FSH做超排处理 ,观察发情 ,适时进行配种 ,于配种后 7d进行非手术法采卵。对照组只是用纯化 FSH做超排处理。参试牛于不同生理时期颈静脉采血 ,分离血清 ,用放免法测定各生理时期血清中 FSH、LH、E2 、P4水平。结果表明 :( 1 )抑制素主动免疫可以提高牛超排效果 ,试验组头均胚胎数和头均可用胚胎数分别比对照组增加 2 .4枚和 1 .4枚 ( P<0 .0 5) ;( 2 )抑制素主动免疫可提高母牛血浆中 FSH水平 ,从而促使卵泡发育 ,增加胚胎数和可用胚胎数 ;( 3)在首免后到加免前 ,抑制素主动免疫对 LH、E2 。
Through active immunization of Inhibin(IB) antigen from porcine seminal plasma and super ovulation of allochthonous purified FSH,the effect of IB active immunization on cow was studied and its mechanism was also further ascertained by examination of relative hormone concentrations in serum at different physiological periold.12 Hostein cows and 12 Xiza young cows of 3 5 years old (postpartum 50 d and above),were selected and divided into the experimental group and control group randomly according to the estrum order.The experimental group were treated with purified FSH for super ovulation on 6 d of the secondary immunization by IB emulsified in incomplete Fowler′s adjuvant which has been immunied on previous 20 d by IB emulsified in complete Fowler′s adjuvant.After super ovulation was inseminated timely,non operational ovum cellecting was done on 7 d of insemination.The cows in the control group were only treated with purified FSH.Blood was sampled from jugular vein at different physiological period and the serum concentration of FSH,LH,E 2 and P 4 were determined by RIA.The results were as follows:(1)The active immunization of IB could improve super ovulation,the number of embryos and the number of embryos available in the experimental group were more 2.4 and 1.4 than that of control gruop (P<0.05);(2)The number of embryes and the number of embryos available could be increased by active immunization of IB indicating that it may increase the concentration of FSH in serum and promote the follicular development;(3)The IB active immunization showed no effect on the LH,E 2 and P 4 secretion prior to the secondary immunization.
