摘要
目的:研究心水通胶囊水提物的细胞毒理学作用。方法:水煎煮法提取心水通胶囊中水溶性成分,不同浓度干预缺氧诱导的人主动脉内皮细胞,并与相同浓度梯度呋塞米组对照。采用MTF比色法、酶学方法以及病理形态学等方法,分别检测细胞活性、培养液中LDH、MDA和SOD含量以及细胞病理形态变化。结果:在0.5-1.5mg/mL浓度之间,心水通胶囊组比缺氧模型组细胞存活率增明显增高(P〈0.05)。在该浓度区间,细胞培养液中LDH和MDA活力降低,而SOD的活力增强,且该作用呈药物浓度依赖性;心水通胶囊组细胞形态正常,贴壁生长良好,未见明显细胞毒性作用。呋塞米组LDH和MDA活力随浓度而增高,而SOD活力和细胞活率降低,细胞毒性也增加。结论:心水通胶囊对缺氧诱导的细胞损伤具有一定保护作用,细胞毒性小。
Objective:To study the water extracts of Xinshuitong Capsule' s toxicity on human aortic endothelial cells. Methods : Use water decoction methods to extract the capsule' s water - soluble components. Oxygen - starved cells were treated with different concentrations of the extracts, and parellel experiments were performed with furosemide. MTI', en- zymology and pathological morphology were applied to test the activity of cells, LDH, MDA and SOD in the medium, and cell pathological changes, respectively. Results : Between 0.5 ~ 1.5 mg/mL, the extracts treating cells showed a dose - dependent and significant increase in cell activity compared with those in the hypoxia model group ( P 〈 0.05 ). Meanwhile, LDH and MDA reduced, but SOD increased in the cell medium, and this effect was also related to the con- centration dependence. The extracts treating cells showed normal in shape and good growth, without clear cell toxic effect in variable concentrations. In Furosemide group we found the increase in LDH and MDA, but decrease in SOD, and the cell toxicity also exacerbated in a dose - dependent manner, Conclusion : Xinshuitong capsule shows a protective effect on hypoxia- induced cells with little cell toxicity.
