摘要
准确测定1,3-二油酸-2-棕榈酸三酰甘油(OPO)的含量,建立了一种以高效液相色谱结合银离子交换柱(Silver-ion HPLC)的测定方法。以ChromSpher 5 Lipid(25 cm×4.6 mm,5μm)银离子交换柱通过HPLC正相系统分离三酰甘油,配合蒸发光散射检测器(ELSD),以毒性较小的二氯甲烷和丙酮为流动相。流速0.6 mL/min,柱温30℃。漂移管温度30℃,空气压力0.35 MPa。在此条件下,1,3-二油酸-2-棕榈酸三酰甘油和1,2-二油酸-3-棕榈酸三酰甘油两种同分异构体基本达到分离,样品出峰完整,峰形良好。OPO色谱峰面积的自然对数与浓度的自然对数呈良好的线性关系(R2=0.998 8)。用该方法测定了母乳脂肪、实验室合成结构油脂样品、商品结构油脂、棕榈油和山茶油中OPO的含量。结果表明该方法的灵敏度高、重现性良好。
A method using Silver- ion HPLC for accurate analysis of 1,3 -dioleoyl -2 -palmitoylglycerol (OPO) has been established here in the paper. Separation and quantification of OPO by ChromSpher 5 Lipid column (25 cm ×4.6 mm,51μm)combined with ELSD using relatively less toxic dichloromethane and acetone as eluents. Flow rate was 0.6 mL/min;column temperature was 30 ℃. The drift tube temperature was 30 ~C and air pressure was 0.35 MPa. Under this condition, 1,3 - dioleoyl - 2 - palmitoylglycerol and 1,2 - dioleoyl - 3 - palmitoylglycerol can be basically separated with a integrity and good shape of peaks The natural logarithm of OPO peak area have a good linear relationship with the natural logarithm of the concentration (R2 = 0. 998 8 ). RSD of precision and recovery were good. The OPO content of human milk fat, OPO sample synthesized in our lab, OPO structured lipid sample, palm oil and camellia oil were analyzed by this method which could be confirmed as high precision repeatability and accuracy.
