摘要
目的观察丙泊酚对大鼠心肌缺血再灌注损伤时凋亡蛋白Bax、Bcl-2表达的影响。方法雄性Wistar大鼠24只,随机分为3组:(n=8),即心肌缺血再灌注组(CI组)、心肌缺血再灌注+丙泊酚组(CP组)、假手术组(CC组)。CI组和CP组采用结扎左冠状动脉前降支30 min再灌注2 h的方法制备心肌缺血再灌注损伤模型。CP组在缺血前10 min开始静脉泵注丙泊酚6 mg.kg-1.h-1至再灌注2 h结束;CC组仅穿线不结扎。再灌注结束后,采用全自动生化分析仪测定大鼠血清乳酸脱氢酶(LDH)、肌酸激酶(CK)水平,免疫组化法测定心肌Bax和Bcl-2表达水平。结果 CI组与CC组相比,LDH、CK水平升高,Bcl-2、Bax表达上调,Bcl-2/Bax比例下降(P<0.05);与CI组比较,CP组LDH、CK水平降低,Bax表达下调,Bcl-2表达上调,Bcl-2/Bax比例升高(P<0.05)。结论大鼠心肌缺血再灌注损伤时丙泊酚可上调抗凋亡蛋白Bcl-2表达、下调凋亡前蛋白Bax的表达。
Objective To investigate the effects of propofol on Bax and Bcl-2 of myocardium ischemia-reperfusion (I/R) injury in rats,and its mechanism.Methods Twenty-four male Wistar rats were randomly divided into three groups (n =8):I/R group (CI),propofol group (CP),and sham group(CC).The cardiac I/R was established by ligaturing left anterior descending coronary artery (LAD) for 30 minutes and then reperfusing for 2 hours in groups CI and group CP.Group CC was just threaded without being ligatured.Group CP was pumped with propofol 6mg · kg-1 · h-1 from 10 min before ischemia to the end of reperfusion.The serum lactate dehydrogenase (LDH) and creatine kinase (CK) were measured after reperfusion.Immunohistochemistry was used to detect the expression of Bax and Bcl-2.Results Compared with group CC,the content of LDH and CK in group CI increased,Bcl-2,Bax increased (P < 0.05),and Bcl-2/Bax ratio decreased (P < 0.05).Compared with group CI,the content of LDH and CK in group CP increased,Bax decreased,Bcl-2 increased,and Bcl-2/Bax ratio increased (P < 0.05).Conclusion Propofol could up-regulate anti-apoptosis protein Bcl-2 and down-regulate proapoptotic protein Bax expression in rats during ischemia/reperfusion injury.
