宫颈癌细胞增殖与组蛋白去乙酰化酶2表达关系分析 被引量:1

Relationship between cervical carcinoma cells proliferation and histone deacetylase-2 expression

摘要 目的:观察拟利用siRNA技术沉默组蛋白去乙酰化酶2(HDAC2)表达其对宫颈癌Hela细胞增殖能力的影响。方法:订购santacruz公司化学成的siRNA,利用lipofectamine2000(脂质体2000)分别转染HDAC2-siRNA或control siRNA到Hela细胞,采用Western blot方法检测HDAC2转染后的表达,挑选干扰效率;用MTT比色法检测实验组和对照组Hela细胞转染后增殖活力的变化;流式细胞仪分析转染48 h两组细胞周期的差异,平板克隆形成实验检测HDAC2 siRNA对Hela细胞增殖的抑制作用。结果:实验组和对照组相比,MTT法检测72 h和96 h增殖倍率明显下降(P<0.01);G0/G1期比例增加,S期细胞、G2/M期细胞比例下降(P<0.05);siRNA-HDAC2组(105±5.5)比阴性对照组(145±3.8),细胞形成的集落数显著减少(P<0.01)。结论:siRNA沉默HDAC2的表达,明显降低了宫颈癌细胞的增殖活力,为肿瘤治疗提供了理论基础。 Objective: To observe the effect of silence expression of histone deacetylase - 2 (HDAC) by siRNA technique on the proliferation of cervical carcinoma Hela cells. Methods: siRNA was synthetised by Santacruz cooperation. HDAC2 - siRNA and control siRNA was transfected into Hela cells. Expression of HDAC2 was detected by Western blot. Hela cell proliferation activity was measured by MTI'. Cell cycle was measured by flow cytometry. Inhibition effect of HDAC2 siRNA on Hela cell was measured by Colony formation assay. Results:Compared with control group, cell proliferation rate was reduced at 72 and 96 h (P 〈 0. 01 ), cell ratio in GO/G1 stage was increased, in S stage and G2/M stage was decreased (P 〈 0. 05 ), colonies were decreased (P 〈 0. 01 ) in observation group. Conclusion:Silence the expression of HDAC2 by siRNA can decrease the proliferation of cervical carcinoma cells, provide theory base for cancer therapy.
