摘要
目的 评价牙龈成纤维细胞、牙周膜细胞对白细胞介素 8(interleukin 8,IL 8)的趋化反应。方法 将体外培养第 6代的牙龈成纤维细胞和牙周膜细胞 (2 5× 10 8个 /L)分别加入Transwell细胞培养室的上室 ,在下室分别加入 10 -10 、10 -9、10 -8及 10 -7mol/L的IL 8。于 37℃ ,95 %空气、5 %CO2下培养 2 4h。光镜下计数滤膜下侧面的细胞数。结果 IL 8在 10 -9~ 10 -7mol/L浓度范围内能增强牙龈成纤维细胞的趋化作用 ,并呈浓度依赖性增加 ;而在相同浓度范围内 ,牙周膜细胞趋化移动的数量并未增加。结论 IL 8参与了调节牙龈成纤维细胞的趋化运动。
Objective To evaluate the chemotactic response of gingival fibroblasts and periodontal ligament cells to interleukin 8 Methods The passage 6 cultured gingival fibroblasts and periodontal ligament cells (2 5×10 8 cell/L, respective) were placed in the upper compartment of the Transwell cell culture chamber The lower compartment contained IL 8 with the concentrations of 10 -10 、10 -9 、10 -8 、10 -7 mol/L, respectively Incubation was at 37℃ in an atmosphere of 95% air and 5% CO 2 for 24 h The number of migrated cells were counted under high power fields Results IL 8 at concentrations ranged from 10 -9 ~10 -7 mol/L promoted the chemotatic movement of gingival fibroblasts in a dose dependent manner However, at the same IL 8 concentrations the chemotactic movement of periodontal ligament cells was not elevated Conclusion IL 8 may play its role in regulating chemotaxis of gingival fibroblast
