摘要
研究了消毒时间、不同激素及浓度配比对罗汉果带腋芽茎段消毒、腋芽诱导、继代培养及生根诱导的影响,从而确定在最短周期内繁殖罗汉果无菌苗的最佳组培快繁方案。结果表明,0.1%升汞最佳消毒时间为8 min,除菌率可达58%;芽诱导培养基的最佳激素组合为6-BA 0.5 mg/L+NAA 0.05 mg/L,诱导率为97.5%;继代培养基最佳激素组合为6-BA 0.5 mg/L+IBA 0.1mg/L+GA30.1 mg/L,增殖率为9;根诱导培养基最佳激素组合为IBA 1.0 mg/L,生根率为98%。GA3能有效促进新芽的分化、增殖和生长。
Aimed at establishing an optimum scheme of tissue culture and rapid propagation to propagate aseptic seedlings of Siraitia grosvenorii in the shortest period, the effects of sterilizing time, different phytohormones and proportions on the explants steri- lization of stem segment with axillary bud, axillary bud induction, multiplication medium and root induction were investigated. The results indicated that explants sterilized in 0.1% aqueous mercuric chloride for 8 min had the best effect and the rate of bacteria and sterilization reached up to 58%. The optimum hormone combination of culture medium on bud induction was 6-BA 0.5 mg/L + NAA O. 05 mg/L and the induction rate was 97.5% under this condition. The optimum hormone combination of culture medium on multi- plication was 6-BA 0.5 mg/L + IBA O. 1 mg/L + GA3 0.1 mg/L and the growth rate could be up to 9. The optimum hormone combi: nation of culture medium on rooting was IBA 1.0 mg/L and the rooting frequency was 98%. GA3 could actually accelerate the differ- entiation, proliferation and growth of adventitious buds.
