摘要
目的探讨在结直肠癌中δ-catenin表达的意义及其与结直肠癌细胞增殖活性的相关性。方法利用免疫组织化学Elivision^(TM)plus法检测结直肠癌组织芯片中120例结直肠癌组织中δ-catenin和Ki-67的表达情况,利用Western blot法检测30例正常结直肠组织和结直肠癌组织中δ-catenin的表达水平。结果结直肠癌组织中δ-catenin的阳性表达率为65.83%(79/120),显著高于正常结直肠组织(P<0.05)TNMⅢ/Ⅵ期的结直肠癌组织中δ-catenin的阳性表达率为78.05%(32/41),明显高于Ⅰ/Ⅱ期结直肠癌组织中的阳性表达率59.49%(47/79)(χ~2=4.132,P=0.042);低分化的结直肠癌组织中δ-catenin阳性表达率为82.14%(23/28),明显高于高、中分化结直肠癌组织中的阳性表达率60.87%(56/92)(χ~2=4.319,P=0.038);δ-catenin在有淋巴结转移的结直肠癌中的表达率为77.05%(47/61),明显高于无淋巴结转移的结直肠癌中的阳性表达率54.24%(32/59)(χ~2=6.939,P=O.008)结直肠癌组织中δ-catenin高表达同结直肠癌细胞的高增殖活性(用Ki-67标记)呈显著正相关(r=0.334,P<0.01)。结论δ-catenin高表达很可能参与了结直肠癌的发生、发展过程。
To investigate the expression and significance of δ-catenin in colorectal cancer (CRC), and explore the correlation between δ-catenin expression and cell proliferation in colorectal cancer. Methods The expressions of δ-catenin and Ki-67 in 120 specimens of colorectal cancer were detected by immunohistochemistry in tissue microassay. The expression of δ-catenin was also determined in 30 paired colorectal cancer and normal colorectum by Western blot. Results Overexpression of δ-catenin was detected in 65.83% (79/120) of the 120 human colorectal can- cer tissues, which was significantly higher than that in normal colorectum. The positive expression of 8-catenin was higher in advanced TNM stages ( Ⅲ+Ⅳ ) of colorectal cancer ( 78.05%, 32/41 ), compared to lower stages ( Ⅰ + Ⅱ ) (59.49%, 47/79 ) (x2=4.132, P = 0.042). The positive expres- sion of δ-catenin was higher in poor differentiated colorectal cancer (82.14%, 23/28), compared to well/moderate differentiated colorectal cancer (60.87%, 56/92) ( X2=4.319, P = 0.038). The positive expression of δ- catenin was higher in colorectal cancer with lymph node metastasis (77.05%, 47/61 ), compared to colorectal cancer without lymph node metastasis (54.24%, 32/59) ( X2=6.939, P= 0.008). In addition, we also found that the overexpression of δ-catenin was positively correlated with proliferation of colorectal cancer which marked by Ki-67 staining in colorec- talcancer (r =0.334, P 〈 0.01 ). Conclusion δ-catenin overexpression might be involved in the initiation and progression of colorectal cancer.
