人血浆中盐酸美沙酮HPLC方法的建立

摘要 目的建立人血浆中盐酸美沙酮的HPLC分析方法。方法采用液-液萃取方法将人血浆中的盐酸美沙酮分离出来,利用C18柱,乙腈-0.02mol/L的磷酸二氢钾溶液(50∶50),2mol/L的氢氧化钠调pH至5.5为流动相,检测波长220nm的色谱条件测定人血浆中盐酸美沙酮。结果盐酸美沙酮在1-100μg/mL的范围内与其峰面积线性关系良好(r=0.9743)。结论本法所建HPLC法操作简便,重复性好,可作为人血浆中盐酸美沙酮的分析测定方法。 Objective To establish the method to determine the content of methadone in the human plasma. Methods Liquid-liquid extraction method is used to seperate methadone from human plasma, using column stuffed with C18, the mobile phase is mixed with acetonitrile of 0.02mol/L and potassium dihydrogen phosphate solution 2mol/L of phosphoric acid or sodium hydroxide of 2mol/L regulated pH to 5.5, the detection walvelength is 220 nm, apply above chromatographic condition to determine the content of methadone. Result Methadone hydrochloride have good linear relationship between peak area in 1-100μg/mL(r=0.9743). Conclusion The HPLC method is simple, and have good repeatability. It can be used as the content determination of methadone in human plasma.
