摘要
目的以活性维生素D3(1,25(OH)2D3)干预抗胸腺细胞抗体诱导的大鼠肾炎模型,观察不同时间点大鼠24h尿蛋白定量水平变化及1,25(OH)2D3对大鼠血肌酐、尿素氮和血钙的影响。方法将45只清洁级雄性SD大鼠随机分为对照组、肾炎模型组、1,25(OH)2D3干预组(干预组)各15只,肾炎模型组和干预组大鼠均经尾静脉一次性注射单克隆抗胸腺细胞抗体,造模成功后用1,25(OH)2D3干预干预组大鼠,分别于干预后第1、3、7、14、21天各选取3只大鼠收集24h尿液并采集动脉血,检测24h尿蛋白定量、血肌酐、尿素氮和血钙水平。结果肾炎模型组在注射单克隆抗胸腺细胞抗体后第1天即产生大量尿蛋白,第3天达高峰,至第21天恢复正常;干预组尿蛋白水平在干预后第1、3、7、14天均明显低于肾炎模型组(P<0.05),且在第14天恢复至正常水平;3组大鼠不同时间点血肌酐、尿素氮和血钙水平比较差异均无统计学意义(P>0.05)。结论 1,25(OH)2D3可降低抗胸腺细胞抗体诱导的肾炎模型大鼠尿蛋白水平,且对肾功能和血钙水平无影响。
Objective To observe the changes of 24-hour urine protein at different time points and the effect of 1,25(OH)2D3 on serum creatinine, urea nitrogen and blood calcium in nephritis rat models induced by anti Thyl monoelonal antibody intervened with 1,25 (OH)2 D3. Methods Forty-five male SD rats were randomly divided into three groups: control group, nephritis group and 1,25 (OH)2D3 group, with 15 rats in each group. Nephritis group and 1, 25 (OH)2 D3 group were intravenously injected anti-Thyl monoclonal antibody once via tail vein. After the nephritis rat models were successfully established, the rats in 1,25 (OH)2 D3 group were intervened by 1,25 (OH)2 D3. The samples of 24-hour urine and arterial blood were collected by day 1, 3, 7, 14 and 21 after intervention to detect 24 hour urinary protein excretion, renal function and electrolyte levels. Results Massive urinary protein was induced in nephritis group by day 1 after injection of anti Thyl monoclonal antibody, reached peak by day 3, and returned to normal hy day 21. The urinary protein levels at different time points were lower in 1,25(OH)2D3 group than those in nephritis group (P〈0.05), and turned to normal by day 14. There were no significant differences in serum ereatinine, urea nitrogen and blood calcium among three groups (P〉0.05). Conclusion 1,25 (OH)2D3 can significantly reduce urinary protein level in nephritis rat models induced by anti-Thyl monoclonal antibody, and has no influence on renal function and blood calcium.
