脑蛋白水解物注射液对脑缺血再灌注大鼠的神经保护作用研究 被引量:7

摘要 目的研究脑蛋白水解物注射液对大鼠脑缺血再灌注损伤的保护作用。方法采用线栓法制作大鼠大脑中动脉闭塞再灌注(MCAO/R)模型,经尾静脉注射脑蛋白水解物注射液300、600、1200μL,于缺血再灌注24h后进行神经功能评分和提高躯体摆动实验(EBST),测定脑匀浆上清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、还原型谷胱甘肽(GSH)含量、一氧化氮合酶(NOS)活性等。结果与模型组比较,脑蛋白水解物注射液高、中、低剂量治疗组神经功能评分和对侧旋转次数水平降低(P<0.05);脑匀浆上清SOD活性和GSH含量明显升高,MDA含量显著降低(P<0.05);总NOS、诱导型NOS(iNOS)、构建型NOS(cNOS)活性差异有统计学意义(P<0.05);上述各指标,中、高剂量组与低剂量组之间比较差异有统计学意义(P<0.05),而中等剂量组和高剂量组间比较无统计学意义(P>0.05)。结论脑蛋白水解物注射液可保护MCAO/R模型大鼠神经功能,该作用与脑蛋白水解物注射液提高模型大鼠脑组织的抗氧化能力、减少脑缺血再灌注所致的氧化性损伤有关,脑蛋白水解物注射液达到一定剂量后其抗氧化作用不再明显提高。 Objective To investigate the neuroprotective effect of Cerebrotein Hydrolysate Injection on ischemia-reperfusion injury in rats.Methods Intraluminal thread methods were applied to establish the middle cerebral artery occlusion reperfusion model (MCAO/R) in rats.Cerebrotein Hydrolysate Injection was injected into the tail vein (300 μL,600 μL,1200 μL).The neurological score and elevated body swing test (EBST) were taken at 24 h after ischemia-reperfusion.The activity of superoxide dismutase (SOD),nitric oxide synthase (NOS),content of malondial dehyde (MDA) and glutathione (GSH) in brain homogenate supernatant were determined.Results Compared with the model group,the neurological function scores and the contralateral of swing numbers of the Cerebrotein Hydrolysate Injection treatment group were significantly reduced (P<0.05);the SOD activity and GSH content of brain homogenate supernatant were significantly increased; MDA content were significantly reduced (P< 0.05).There were significant differences of total NOS,induced type NOS (iNOS) and building type NOS (cNOS) activity between model and treatment group (P<0.05).There were significant differences between the medium,high dose group and the low dose group (P<0.05),but there was no significant statistical significance between the medium and high dose group (P > 0.05).Conclusions Cerebrotein Hydrolysate Injection can protect neural function of MCAO/R model rat,the mechanism may be related to the improving of oxidation resistance ability and reducing cerebral ischemia reperfusion injury caused by oxidizing.After Cerebrotein Hydrolysate Injection reaches a certain dose,its antioxidation function no longer obviously improved.
