摘要
目的观察二甲双胍(MF)对去势大鼠血清中TNF-α,IL-6及瘦素含量的影响,探讨MF对去势大鼠瘦素抵抗的改善作用和免疫学机制。方法选取Wistar雌性去势大鼠30只,随机分为去势模型组、MF低剂量组(MFL)和MF高剂量组(MFH),每组10只,另选健康Wistar雌性大鼠10只为假手术对照组,用药组分别给予MF 135mg·kg^(-1)·d^(-1),270mg·kg^(-1)·d^(-1),去势模型组和假手术对照组分别给予等容量生理盐水,灌胃,连续给药42d。采用ELISA法,测定大鼠血清TNF-α及IL-6含量;采用RIA法测定大鼠血清瘦素水平。结果与假手术对照组比较,去势大鼠血清TNF-α,IL-6及瘦素含量显著升高,同时体重明显增加(P<0.05);与去势组比较,二甲双胍高剂量组血清TNF-α,IL-6及瘦素水平则明显降低,体重亦显著降低(P<0.05),而二甲双胍低剂量组无显著变化(P>0.05)。结论 MF可显著降低去势大鼠血清瘦素含量,降低去势大鼠体重,其机制与调节去势大鼠血清TNF-α及IL-6水平有关。
Objective To observe the effects of metformin on serum concentration of leptin ,TNF-αand IL-6 in ovariectomized rats .Methods Thirty OVX rats were randomly divided into three group ( 10 rats in each group ):OVX model group,low dose MF[135mg/(kg· d),ig] group,high dose MF[270mg/(kg· d),ig] group.Ten healthy rats were chosen as the sham control group ,ig,for 42 days.The low dose MF group was treated with MF in 135mg/(kg· d),and the high dose MF group in 270mg/(kg· d).Other groups were treated with saline solution in the same capaci-ty.The serum concentration of TNF-αand IL-6 was measured with ELISA ,and leptin level was measured with RIA .Re-sults Compared with sham rats ,the serum concentration of TNF-α,IL-6 and leptin in OVX mice were significantly in-creased,as like as the weight of OVX rats (P〈0.05).Compared with OVX rats,the serum concentration of TNF-α,IL-6 and leptin in rats treated with the high dosage MF was obviously decreased ,just like the weight in OVX rats(P〈0.05). However,the low dose MF group was short of significant diversities .Conclusion MF could reduce the level of serum leptin and weight ,and the mechanism is related primily to the levels of serum TNF-αand IL-6 in OVX rats.
